Synthesis and Evaluation of a Library of Alternating Amphipathic Copolymers to Solubilize and Study Membrane Proteins

Adrian H. Kopf et al.

Summary: In this study, a library of well-defined alternating copolymers of SMA analogues was designed and synthesized, revealing important polymer properties for membrane protein solubilization and stability. Remarkably, halogenation of the SMA copolymers significantly improved solubilization efficiency, while substituents on the copolymer backbone enhanced resistance to Ca2+.

Mechanism of gating and partial agonist action in the glycine receptor

Jie Yu et al.

Summary: Using electrophysiology and cryo-EM reconstructions, this study investigated the mechanism of partial agonist action on neurotransmitter receptors, revealing new conformational states along the receptor reaction pathway. The results demonstrated that partial agonists can occupy closed channel states bound by agonists, affecting the receptor channel's open probability.

Benchmarks of SMA-Copolymer Derivatives and Nanodisc Integrity

Giacomo M. Di Mauro et al.

Summary: The study compares copolymers and copolymer-lipid nanodiscs obtained by SMA-EA with a set of anionic XIRAN copolymer formulations commercially available. The results show significant alterations in critical micellar concentration (c.m.c.) of each copolymer in the presence of lipids and confirm an equilibrium between nanodisc-bound and free/micellar copolymer chains. These findings can be utilized to optimize studies involving special copolymers, simplifying applications and expanding the scope of polymer-based nanodiscs.

Synthesis, Characterization, and Nanodisc Formation of Non-ionic Polymers

Thirupathi Ravula et al.

Summary: A family of non-ionic amphiphilic polymers synthesized by hydrophobic functionalization of fructo-oligosaccharides/inulin has been reported to improve the interaction between lipid nanodiscs and membrane proteins. These polymers show stability against pH and divalent metal ions, as well as efficient extraction of membrane proteins.

Structural biology of endogenous membrane protein assemblies in native nanodiscs

Chanelle J. Brown et al.

Summary: The development of amphiphilic copolymers has allowed the solubilization of integral membrane proteins into native nanodiscs, providing insights into their structures and functions. This breakthrough has established biological membrane:protein assemblies as fundamental units of cellular membranes, and various methods including cryo-EM and NMR spectroscopy have been utilized for the characterization of transmembrane proteins within these nanodiscs.

Solid-State NMR Study to Probe the Effects of Divalent Metal Ions (Ca2+ and Mg2+) on the Magnetic Alignment of Polymer-Based Lipid Nanodiscs

Thirupathi Ravula et al.

Summary: Divalent cations, especially Ca2+ and Mg2+, have a significant impact on the formation and magnetic alignment properties of polymer-based lipid nanodiscs. The interaction of these cations with lipid headgroups can promote the stacking up of nanodiscs, resulting in enhanced magnetic alignment. By optimizing the temperature and concentration of divalent metal ions, the optimal alignment of nanodiscs in the presence of an applied magnetic field can be achieved, which is crucial for various applications and structural studies using NMR.
