Summary: The new cyclophilin inhibitor C31 effectively inhibits cardiac mitochondrial permeability transition pore (mPTP) opening in vitro, showing potential for cardioprotection. However, the lack of efficacy of C31 and cyclosporine A after systemic administration highlights the challenges in targeting myocardial mitochondria in vivo, which should be considered in cardioprotective strategies.
