Quantitative comparison of different fluorescent dye-loaded nanoparticles

Guangze Yang et al.

Summary: Labeling nanoparticles with fluorescent dyes is a common method to study their cell uptake and biodistribution for drug delivery applications. However, the assumption that fluorescence intensity correlates with the amount of nanoparticles taken up by cells may not always hold true due to factors like dye dispersion, quenching, and material shading. This study shows that both hydrophobic and hydrophilic dye-labeled nanoparticles can suffer from dye quenching, making quantitative comparison of cell uptake challenging. The proposed solution aims to enable direct comparative studies of dye-labeled nanoparticles for better design and evaluation in drug delivery.

Implications of Quenching-to-Dequenching Switch in Quantitative Cell Uptake and Biodistribution of Dye-Labeled Nanoparticles

Guangze Yang et al.

Summary: This study compared two types of dye-labeled nanoparticles and found that nanoparticles with dye molecules in a solid state undergo dye quenching, while those in a soluble state do not. To standardize the comparative study, the proposed solutions include using lower dye loading or utilizing medium analysis for quantifying cell uptake of nanoparticles. Valuable insights into selecting valid quantification methods for bio-nano studies are provided.

Particle Integrity and Size Effect on the Journey of Polymeric Nanocarriers in Zebrafish Model and the Correlation with Mice

Jinsong Tao et al.

Summary: The study investigates the biofate correlation of different-sized poly(caprolactone) nanocarriers in zebrafish and mice, showing that PEGylated PCL NCs have higher integrity in both models. Small PEG-PCL NCs have longer circulation time, while large ones have higher macrophage sequestration in both species. The findings suggest that 7 dpf zebrafish are suitable for in vivo screening of nanocarriers and predicting their performance in rodents.
