Summary: The study reveals that stress fibers can form without pre-existing actomyosin bundles, giving rise to structures known as cortical stress fibers, which are embedded in the cell cortex and assemble preferentially underneath the nucleus. The assembly process is orchestrated by non-muscle myosin II pulses, leading to the reinforcement of nascent focal adhesions and subsequent stabilization of the cortical stress fibers. These findings demonstrate a new mechanism for de novo generation of stress fibers from the actin cortex, highlighting the role of stochastic myosin pulses in the assembly of functional actomyosin bundles.
