Binding pathway determines norepinephrine selectivity for the human β1AR over β2AR

Summary: Beta adrenergic receptors mediate physiological responses to catecholamines epinephrine and norepinephrine, and norepinephrine shows higher affinity for beta(1)AR. Crystal structure analysis revealed identical catecholamine-binding pockets but differences in extracellular vestibules between beta(1)AR and beta(2)AR, influencing norepinephrine binding path and affinity.

Exploring the putative mechanism of allosteric modulations by mixed-action kappa/mu opioid receptor bitopic modulators

Summary: The modulation and selectivity mechanisms of seven mixed-action kappa opioid receptor (KOR)/mu opioid receptor (MOR) bitopic modulators were explored in this study. It was found that the 'message' moiety shared the same binding mode with the orthosteric site of the KOR and MOR, while the 'address' moiety bound with different subdomains of the allosteric site. The 3-hydroxy group on the phenolic moiety induced selectivity to the KOR over the MOR.

Summary: The efficacy of alpha-2 adrenergic receptor (α2AR) agonists in relieving chronic neuropathic pain varies greatly among individuals, and is influenced by the timing of treatment post-establishment of neuropathic pain and the activity of regulator of G-protein signaling protein 4 (RGS4). Increased plasma membrane expression of RGS4 may be associated with reduced efficacy of α2AR agonists in a model of persistent neuropathic pain.

Summary: Allosteric modulators (AMs) of G-protein coupled receptors (GPCRs) present desirable drug targets due to their potential to produce fewer on-target side effects and improved selectivity compared to orthosteric drugs. Peptides and proteins, deriving from endogenous protein-protein interactions, intramolecular receptor contacts, endogenous peptides, and exogenous libraries, serve as underappreciated sources for identifying AM leads, offering advantages such as high affinity and bioactivity along with disadvantages like poor metabolic stability. Peptidomimetics combine the advantages of both peptides and small molecules, mimicking peptide chemical features responsible for bioactivity while enhancing druggability.

Experimental evidence shows that a specific mu-PAM, BMS-986122, can enhance the potency of endogenous opioid peptides and produce antinociceptive effects in mouse models of acute and inflammatory pain. This innovative approach to pain management may offer a safer alternative to conventional opioid medications.

Controlling the selectivity of aminergic GPCR ligands from the extracellular vestibule

Summary: Recent structural studies have identified several allosteric sites for pharmacological intervention in addition to the orthosteric binding pockets of GPCRs. Through a virtual screening protocol, new bitopic compounds targeting specific receptor profiles were designed. The designed compounds showed biased selectivity towards the desired receptor targets and successfully addressed limitations of known drugs.

Fragment evolution for GPCRs: the role of secondary binding sites in optimization

Evaluation of 13,000 extensions for beta(1)- and beta(2)-adrenergic receptors resulted in the synthesis and testing of 112 bitopic molecules, confirming the positive contribution of the secondary binding pocket to potency and selectivity optimizations.
