Cannabidiol selectively inhibits the contraction of rat small resistance arteries: Possible role for CGRP and voltage-gated calcium channels

Amna C. Mazeh et al.

Summary: This study demonstrates that cannabidiol inhibits the contraction of small resistance arteries through a combination of mechanisms involving CGRP and L-type calcium channels, while large arteries are insensitive to cannabidiol. Moreover, cannabidiol has little effect on bronchi, atria, and hemidiaphragm, but can attenuate maximum contractions in vasa deferentia. The high sensitivity of resistance arterial circulation to cannabidiol in the clinical range may present a therapeutic opportunity in peripheral vascular disease.

The ADAMTS13-von Willebrand factor axis in COVID-19 patients

Ilaria Mancini et al.

Summary: Severe COVID-19 patients exhibit an increased risk of thromboembolic events, with evidence of microthrombosis in the lungs of deceased patients. The study found a significant alteration of the VWF-ADAMTS13 axis in COVID-19 patients, with an elevated VWF:Ag to ADAMTS13 activity ratio strongly associated with disease severity, enhancing the hypercoagulable state and risk of microthrombosis in these patients.

Summary: Studies suggest that CBD may have cardioprotective properties in hypertensive conditions, but could have adverse effects on cardiac structure and function in normotensive hearts. Caution should be exercised when using CBD as a therapeutic drug.

Food effects on the formulation, dosing, and administration of cannabidiol (CBD) in humans: A systematic review of clinical studies

Lucy H. Silmore et al.

Summary: CBD is increasingly being sought after as a treatment for various ailments, but its bioavailability and pharmacokinetics are not well understood, and proper dosing schemes have not been adequately developed for clinical use. Research has shown that CBD has higher bioavailability when taken with food, and the administration of Epidiolex in the fed state leads to lower interindividual variability in pharmacokinetics. The main excipient in oral CBD formulations, refined sesame seed oil, is a major contributor to the dose-dependent variations in CBD pharmacokinetics.

Guidelines for the measurement of vascular function and structure in isolated arteries and veins

Camilla F. Wenceslau et al.

Summary: The measurement of vascular function in isolated vessels provides important insights into the cardiovascular system's structure and pathological changes. It has led to the discovery of treatments for cardiovascular diseases and raised concerns about scientific rigor and reproducibility. Therefore, establishing guidelines for evaluating isolated vascular function is necessary to ensure consistent and reliable results.

Summary: The endothelium, a cellular monolayer lining the blood vessel wall, plays a critical role in maintaining multiorgan health and homeostasis. Dysfunction of the vascular endothelium is associated with various diseases and is implicated in severe coronavirus disease 2019. Therapeutic approaches targeting endothelial dysfunction may have potential for treatment of cardiovascular and many other diseases.
