A facile synthesis of diaryl pyrroles led to the discovery of potent colchicine site antimitotic agents

Summary: Three different series of cis-restricted analogues of combretastatin A-4 (CA-4) were synthesized and evaluated for their antiproliferative activity against human cancer cell lines. Compound 11j showed superior intracellular target inhibition, blocking the cell cycle and inducing apoptosis via the mitochondrial pathway at low toxicity for normal cells. In vivo, compound 11j demonstrated significantly reduced tumor growth, even at lower doses, compared to CA-4P.
