Aberrant Gamma-Band Oscillations in Mice with Vitamin D Deficiency: Implications on Schizophrenia and its Cognitive Symptoms

Fine structure analysis of perineuronal nets in the ketamine model of schizophrenia

Summary: Perineuronal nets (PNNs) are highly condensed brain extracellular matrix structures that play a key role in postnatal brain development, cell excitability, and are implicated in brain disorders such as schizophrenia. A new open-source script was developed to quantify structural alterations of PNNs in a schizophrenia rat model. The intensity, size, and shape of PNN units were found to be the major factors determining their variability, with ketamine-treated rats showing differences in these parameters compared to control rats. Multidimensional analysis of PNN units provided a comprehensive morphometric fingerprinting of ECM structure abnormalities in the experimental schizophrenia model.

Perineuronal Nets in the Prefrontal Cortex of a Schizophrenia Mouse Model: Assessment of Neuroanatomical, Electrophysiological, and Behavioral Contributions

Summary: Schizophrenia is a neurodevelopmental disorder with dysregulation of perineuronal nets (PNNs) associated with parvalbumin-positive interneurons (PVs) in the prefrontal cortex (PFC). Using a DISC1 mutation mouse model, it was found that both PNNs and PVs were significantly reduced during development, leading to behavioral and neural activity changes resembling schizophrenia-like symptoms after enzymatic digestion of PNNs. These findings suggest that aberrant development of PNNs and PVs in the PFC can impact neural operations and contribute to behavioral phenotypes in schizophrenia.

PNNs mainly surround parvalbumin (PV)-containing, fast-spiking GABAergic interneurons, which inhibit surrounding neurons and contribute to gamma oscillations important for various cognitive functions.
