Nanopores: a versatile tool to study protein dynamics

Summary: Proteins are essential for cellular functions, but observing their functional cycle involving conformational changes and interactions is challenging. Single-molecule techniques like FRET and force spectroscopies provide insights but have limitations. Electrical nanopore detection offers a wide range of timescales relevant for protein dynamics and may provide new solutions for studying protein function at the molecular level.

Palladium zero-mode waveguides for optical single-molecule detection with nanopores

Summary: Metal film nanopores, known as zero-mode waveguides, prevent light transmission with wavelengths much larger than the pore diameter while allowing molecules to pass through. By using palladium as a metal film material for ZMWs, it is possible to simultaneously detect translocations of individual fluorophores of different colors, enabling high signal-to-noise ratio single-molecule biophysics studies.

Magnetic tweezers have also been developed to study DNA topology and topoisomerase functioning, providing a longer observation window for single biomolecules.

Discovering multiple types of DNA methylation from bacteria and microbiome using nanopore sequencing

Summary: In this study, a tool named nanodisco was developed to successfully apply nanopore sequencing for broadly applicable DNA methylation discovery and reliable methylation detection in bacteria and mouse gut microbiome. Furthermore, the application of DNA methylation for binning metagenomic contigs, associating mobile genetic elements with their host genomes, and identifying misassembled metagenomic contigs was demonstrated.
