Chromosome-level de novo assembly of Coprinopsis cinerea A43mut B43mut pab1-1 #326 and genetic variant identification of mutants using Nanopore MinION sequencing

Yichun Xie et al.

Summary: The homokaryotic Coprinopsis cinerea strain A43mut B43mut pab1-1 #326 is a widely used experimental model for developmental studies in mushroom-forming fungi. A chromosome-level genome assembly of strain #326 was reported, providing improved contiguity and annotations, suitable for genetic, genomic and transcriptomic studies. This study showed the usage of MinION for constructing a fungal reference genome and downstream analyses, offering solutions and parameters for fungal genomic analysis on the MinION sequencing platform.

Ultrastructure and development of acanthocytes, specialized cells in Stropharia rugosoannulata, revealed by scanning electron microscopy (SEM) and cryo-SEM

Ying Yang et al.

Summary: This study investigated the ultrastructure and development of acanthocytes in S. rugosoannulata using SEM and cryo-SEM, revealing three distinct stages of acanthocyte development and the presence of irregular patches and holes on their branches. Environmental nitrogen levels impacted acanthocyte production, but acid medium only delayed initiation of formation without affecting morphology. Acanthocytes of S. rugosoannulata exhibit similar hydrophobicity to mycelia, providing insights into their ecological function and evolution.

Label-Free Comparative Proteomics Analysis Revealed Heat Stress Responsive Mechanism in Hypsizygus marmoreus

Lili Xu et al.

Summary: Heat stress response in Hypsizygus marmoreus involves autolysis and apoptosis of mycelia, mediated by upregulation of cell wall degradation enzymes and inhibition of cell wall synthesis enzymes, and induction of ROS and caspases, respectively. Accumulation of ROS triggers antioxidant defense system activation and MAPK cascade involvement in signal transduction. Additionally, inducible expression of heat shock proteins and upregulation of trehalose synthesis enzymes were found, along with proteasome upregulation to prevent protein misfolding. Energy production enzymes were also upregulated to maintain cellular processes during heat response.

Summary: This study reports the near-complete high-quality genome of Hypsizygus marmoreus and reveals the genetic variation and morphological features relationship through comparative genomics analysis and population classification. Furthermore, a candidate causal gene for melanogenesis in H. marmoreus was identified based on genetic population study, bulked segregant analysis (BSA), and comparative transcriptome analysis.

Effects of drying on the structural characteristics and antioxidant activities of polysaccharides from Stropharia rugosoannulata

Qi Wang et al.

Summary: Different drying methods were used on Stropharia rugosoannulata to investigate the structural characteristics and antioxidant activities of two types of neutral polysaccharides and two types of acidic polysaccharides. Hot-air drying was found to alter the structural configuration and enhance the antioxidant activity of the polysaccharides, with hot-air dried polysaccharides exhibiting stronger antioxidant properties.
