The HDL Proteome Watch: Compilation of studies leads to new insights on HDL function

W. Sean Davidson et al.

Summary: The HDL proteome, identified through various studies using different methodologies across multiple laboratories, provides valuable insights into HDL function. Known functions of consensus HDL proteins extend beyond traditional roles in lipid transport, pointing towards involvement in innate immunity, inflammation, cell adhesion, hemostasis, protease regulation, as well as vitamin and metal binding. Further research is needed to address significant issues identified in these studies.

Lipid and metabolic syndrome traits in coronary artery disease: a Mendelian randomization study

David G. Thomas et al.

Summary: The study assessed the pleiotropic effects of lipid trait genetic variants and conducted multivariable Mendelian randomization (MR) analysis in coronary artery disease. Results indicated that 30% of lipid trait genetic variants have effects on metabolic syndrome traits, yet the associations of LDL-C, HDL-C, TGs, BMI, T2D, and SBP with CAD remained independent and robust to adjustment for directional pleiotropy. MR at loci linked to direct effects on HDL-C and TGs suggested specific causal effects of these factors on CAD.

A Mendelian randomization study of the role of lipoprotein subfractions in coronary artery disease

Qingyuan Zhao et al.

Summary: Recent genetic data has provided important insights into the roles of lipoprotein subfractions and particle sizes in preventing coronary artery disease, with medium HDL particles and small HDL particles showing potential protective effects against CAD. Further investigations are required to fully understand the role of HDL particle size in CAD prevention.

HDL in the 21st Century A Multifunctional Roadmap for Future HDL Research

Summary: Low high-density lipoprotein cholesterol (HDL-C) is associated with adverse lifestyle choices, cardiovascular diseases, inflammatory disorders, and other conditions. Therapeutic strategies targeting HDL-C levels have not been successful, and future focus should be on optimizing HDL function for better outcomes.

Artificial high-density lipoprotein-mimicking nanotherapeutics for the treatment of cardiovascular diseases

Hongliang He et al.

Summary: Despite the efficacy of current therapies in lowering low-density lipoprotein cholesterol to reduce cardiovascular disease risks, there are still significant risks of morbidity and mortality from CVD in the general population. Clinical trials have been conducted to investigate the potential of direct infusion of reconstituted HDL products to reduce coronary events in CVD patients, and there is also a growing focus on developing artificial HDL-mimicking nanotherapeutics for complementary therapeutic strategies for CVD patients beyond traditional approaches.

Changing Perspectives on HDL: From Simple Quantity Measurements to Functional Quality Assessment

Himani Thakkar et al.

Summary: HDL is a heterogeneous group of particles with varying size, density, and composition, but its role in cardiovascular diseases remains controversial. Recent research has shifted focus from HDL quantity to HDL quality, particularly its atheroprotective functions. Future research aims to improve HDL functions by altering its composition to reduce cardiovascular risk.
