Organoid systems to study the human female reproductive tract and pregnancy

Summary: The proper functioning of the female reproductive tract and normal placental development are essential for women's health, but studying the FRT in humans has been challenging due to tool limitations. Recent advances in organoid technology have opened up new avenues for research.

Opposing Wnt signals regulate cervical squamocolumnar homeostasis and emergence of metaplasia

Summary: The transition zones of squamous and columnar epithelia are prone to cancer, often associated with metaplasia. Through mouse models and single-cell RNA sequencing, it was discovered that the cervical epithelial cells in the endo- and ectocervix have distinct origins from stem cells regulated by opposing Wnt signals from the stroma. Additionally, a mouse model of cervical metaplasia showed that the stroma in the endocervix undergoes remodeling and increases expression of the Wnt inhibitor DKK2, promoting the outgrowth of ectocervical stem cells.

The mucosal epithelial lining in various regions of the FRT enables selective transport of gametes and successful transfer of the zygote. Three-dimensional organoid systems from the FRT now offer valuable experimental models for studying normal physiology and disease.
