Methods to investigate intrathecal adaptive immunity in neurodegeneration

Hamilton Oh et al.

Summary: CSF serves as a protective barrier for the brain, with recent studies indicating involvement of T cells in neurodegenerative diseases. A standardized method for long-term preservation of CSF immune cells was presented, along with bioinformatic approaches to identify disease-associated TCRs in patients. These novel methods will enhance understanding of adaptive immunity in the central nervous system.

Differential Effects of Fingolimod and Natalizumab on B Cell Repertoires in Multiple Sclerosis Patients

M. C. Kowarik et al.

Summary: Natalizumab and fingolimod have differential effects on B cell populations in multiple sclerosis, with fingolimod treatment contracting peripheral blood B cells while natalizumab treatment expands them. The clonal overlap between CSF and PB B cells is reduced with natalizumab but remains stable with fingolimod therapy. These findings suggest that the two therapies have distinct impacts on the exchange and clonal expansion of B cells in MS patients.

Summary: Deep immune phenotyping identified a B-cell population associated with multiple sclerosis, with increased levels of B cell-related factors in early disease stages. Soluble signaling molecules related to B cells, including FCRL2 and BAFF, were altered in multiple sclerosis.

The CXCL13/CXCR5-chemokine axis in neuroinflammation: evidence of CXCR5+CD4 T cell recruitment to CSF

Christine Harrer et al.

Summary: The study found a strong correlation between intrathecal CXCL13 elevations and CXCR5+CD4 T cell frequencies in CSF, suggesting recruitment of possible professional B cell helpers to the inflamed CNS, which is important for infection control, chronic inflammation, and CNS autoimmunity.
