Recent trends and developments of PCR-based methods for the detection of food-borne Salmonella bacteria and Norovirus

This study demonstrates that the adverse effects of stem-loop structures on PCR performance are directly correlated with their thermal stability, with stable structures leading to truncated intermediate PCR products. Moreover, endonuclease activity of Taq DNA polymerase can unwind stem-loop structures during PCR extension, providing new mechanistic insights into the complexity of PCR assays.
