Effect of biochemical and biomechanical factors on vascularization of kidney organoid-on-a-chip

Summary: By mimicking shear stress fluid flow and optimizing extracellular matrix conditions, our kidney organoid-on-a-chip system showed more mature podocytes and vascular structures compared to static culture, with increased sensitivity to nephrotoxic drugs. Physiological flow was demonstrated to play a crucial role in maintaining various physiological functions of kidney organoids.

Microvascularized tumor organoids-on-chips: advancing preclinical drug screening with pathophysiological relevance

Summary: The recent advancements in organoids engineering and organ-on-a-chip microfluidic technologies have allowed for the recapitulation of the major functions and structures of microscale human tissue, including tumor pathophysiology. However, challenges still exist in replicating the complexity and heterogeneity of the tumor microenvironment. The integration of these engineering technologies may offer a potential strategy to overcome limitations in reconstituting the perfusable microvascular system of large-scale tumors while conserving their key functional features.

Measuring barrier function in organ-on-chips with cleanroom-free integration of multiplexable electrodes

Summary: TEER measurements can be used in OoCs to estimate barrier properties of tissues or cell layers in a non-invasive manner. This article presents a cleanroom-free fabrication method for integrating platinum electrodes in an OoC, demonstrating the real-time assessment of barrier function. The system's effectiveness is shown by creating gut-on-chip and blood-brain-barrier models, expanding potential applications for monitoring barrier formation in OoCs.
