Mitochondrial membranes modify mutant huntingtin aggregation

Adewale Adegbuyiro et al.

Summary: The study found that mitochondrial membranes play a significant role in influencing the aggregation of the huntingtin protein in Huntington's disease, with the inner mitochondrial membrane having a greater impact. Cardiolipin (CL) was identified as a key player in mhtt aggregation, with different molar ratios affecting fibrillization processes.

Embryo and Its Mitochondria

Pascale May-Panloup et al.

Summary: Mitochondria, present in almost all eukaryotic cells, have their own genome and play crucial roles in oocytes and early embryo development. They are considered as potential biomarkers of oocyte competence and embryo viability, with further studies needed to confirm their significance in reproduction.

Dynamin-related protein 1 inhibition reduces hepatic PCSK9 secretion

Maximillian A. Rogers et al.

Summary: The study demonstrates that inhibition of the mitochondrial fission protein DRP1 can reduce remodeling of specific ER microdomains, promoting mitochondrial fission, autophagy, and enhancing mitochondrial calcium buffering and PCSK9 degradation. The findings suggest a novel role of DRP1 in regulating hepatic proteostasis and mdivi-1 as a potential small molecule PCSK9 inhibitor.

Dominant mutations in MIEF1 affect mitochondrial dynamics and cause a singular late onset optic neuropathy

Majida Charif et al.

Summary: Inherited optic neuropathies are the most common mitochondrial diseases, with neurodegeneration and loss of retinal ganglion cells being key characteristics. Mutations in the MIEF1 gene, which encodes the MID51 protein, have been identified as a cause for a rare form of late-onset progressive optic neuropathy. These mutations disrupt mitochondrial network dynamics without affecting MID51's localization or oligomerization on the outer mitochondrial membrane. Properly regulated mitochondrial dynamics play a crucial role in neurodegeneration.

IFN-β rescues neurodegeneration by regulating mitochondrial fission via STAT5, PGAM5, and Drp1

Emilie Tresse et al.

Summary: The study found that neuronal IFN-beta is crucial for maintaining mitochondrial homeostasis and metabolism by controlling mitochondrial fission, ATP levels, and preventing excessive ROS. Lack of neuronal IFN-beta in a Parkinson's disease model disrupts mitochondrial function, leading to the accumulation of damaged mitochondria and excessive oxidative stress.

Distinct fission signatures predict mitochondrial degradation or biogenesis

Tatjana Kleele et al.

Summary: Mitochondrial fission is a highly regulated process that can impact cell function and health. Depending on the location and manner of division, mitochondria can be divided into two types, one for clearing damaged material and undergoing mitophagy, and the other for mitochondrial proliferation.

Accelerated expansion of pathogenic mitochondrial DNA heteroplasmies in Huntington's disease

Yiqin Wang et al.

Summary: Mitochondrial dysfunction is accelerated in patients with Huntington's disease due to the presence of pathogenic mtDNA heteroplasmies, particularly in lymphoblasts. The expansion of these heteroplasmies is correlated with disease progression and severity, as well as decline in functional capacity. Additionally, elongated CAG repeats in the HTT gene contribute to the age-dependent expansion of pathogenic mtDNA heteroplasmies in HD patients.
