High-resolution multilocus sequence typing for Chlamydia trachomatis: improved results for clinical samples with low amounts of C. trachomatis DNA

Summary: This study improved Bom's MLST scheme by using newly designed primers and stringent conditions, enhancing the detection performance for samples with low concentrations of Chlamydia DNA. A simple scoring scheme and flow chart were developed based on Real-Time PCR analysis values to identify reaction inhibitors and predict amplification success. The new MLST version showed great results in patients attending an STI clinic.

Summary: This article adapts the analysis method of GB-speckles to detect differences in a group of genes for MLST, showing high sensitivity and accuracy as an alternative to classical MLST. The use of s-LASCA imaging of GB-speckles has been demonstrated to be highly efficient, representing significant progress in digital biology and improvements in the bio-digitalization of bacterial DNA.

Genome data vs MLST for exploring intraspecific evolutionary history in bacteria: Much is not always better

Summary: In this study, it was found that genome-based phylogeny and MLST-based phylogeny showed varying levels of incongruence in the phylogeny of each bacterial species analyzed. While most branches in the genome-based trees had high branch support, a significant number of branches also showed high levels of topological incongruence in individual trees. This suggests that high branch supports obtained in genome phylogenies could be influenced by data size, and MLST is a valid method for addressing intraspecific phylogenetic structure. Each bacterial species has its own evolutionary history, which may be impacted to different extents by horizontal gene transfer.

Summary: Chlamydia cases in Guangdong province have been increasing over the past 11 years, with its expansion linked to indicators of social and economic development. Areas with higher male-to-female ratios and greater urbanization are more likely to have higher chlamydia case occurrence. Future control efforts should focus on a population-based approach targeting sexually active adults from diverse economic and migratory backgrounds.
