Epidemiology of Mycobacterium abscessus in England: an observational study

Samuel Lipworth et al.

Summary: This study investigated the transmission of Mycobacterium abscessus in healthcare settings in England by analyzing whole-genome sequencing data and healthcare usage information. The findings showed that there were similar levels of genomic clusters in patients with and without cystic fibrosis, and that person-to-person transmission was not the main mechanism for M abscessus dissemination at a national level in England. The study provides reassurance that the risk of acquisition from other patients in healthcare settings is relatively low and suggests future research should focus on identifying acquisition routes outside of the cystic fibrosis healthcare-associated niche.

A novel DNA chromatography method to discriminate Mycobacterium abscessus subspecies and macrolide susceptibility

Mitsunori Yoshida et al.

Summary: This study identified genetic markers for discriminating MABC subspecies using whole genome sequencing, and developed PCR and DNA chromatography assays for rapid detection. The DNA chromatography method showed high accuracy in identifying macrolide susceptibility in MABC isolates, with a 99.7% agreement rate with WGS-based identification.

Serum GPL core antibody levels are associated with disease activity and treatment outcomes in Mycobacterium avium complex lung disease following first line antibiotic treatment

Kiyoharu Fukushima et al.

Summary: In patients with MAC-LD, serial measurements of GPL core antibody levels were found to be associated with disease activity and treatment outcomes. Patients who achieved treatment success had significantly reduced antibody levels after antibiotic treatment, while those who experienced treatment failure or recurrence did not show significant reductions.

Drug Resistance in Nontuberculous Mycobacteria: Mechanisms and Models

Saloni Saxena et al.

Summary: The review focuses on the rise in infections caused by nontuberculous mycobacteria (NTM) and their resistance to conventional antibiotics, proposing strategies to develop better therapies against them. It suggests accelerating the drug development pipeline by utilizing a combination of computational, laboratory and animal testing methods. The review also highlights the importance of understanding NTM virulence and developing new effective antimicrobial therapies.

Investigation of two cases of Mycobacterium chelonae infection in haemato-oncology patients using whole-genome sequencing and a potential link to the hospital water supply

T. Inkster et al.

Summary: This study investigated two cases of M. chelonae infection in patients and positive water samples in a hospital by conducting water testing, whole-genome sequencing, and implementing infection control measures. The results showed that the patient strains were unrelated to each other, but one patient's isolate was closely related to environmental samples from water outlets, supporting nosocomial acquisition. Discussions on relevant control measures and the potential for chemical dosing of water systems to enhance proliferation of atypical mycobacteria were included.

Mycobacterium abscessus Strain Morphotype Determines Phage Susceptibility, the Repertoire of Therapeutically Useful Phages, and Phage Resistance

Rebekah M. Dedrick et al.

Summary: Mycobacterium abscessus is an opportunistic pathogen with widespread multidrug resistance, making treatment difficult. However, the use of bacteriophages has shown potential as an alternative approach, especially for rough strains. Studying the phage infection profiles of clinical isolates has revealed that colony morphotype is a key indicator of phage susceptibility, leading to the possibility of using small cocktails or single phages for controlling infections.

The global outbreak of Mycobacterium chimaera infections in cardiac surgeryda systematic review of whole-genome sequencing studies and joint analysis

Summary: The international cardiac surgery-associated Mycobacterium chimaera outbreak was likely linked to contamination of LivaNova heater-cooler devices (HCDs) during production, with strain 1.1 causing most infections. Whole-genome sequencing (WGS) revealed genetic relatedness between M. chimaera isolates from patients and LivaNova HCDs, supporting the hypothesis of a point source.

The Antimalarial Mefloquine Shows Activity against Mycobacterium abscessus, Inhibiting Mycolic Acid Metabolism

Giulia Degiacomi et al.

Summary: The study focused on the essential mycolic acids transporter MmpL3 and identified mefloquine as a potential new drug against Mycobacterium abscessus. Mefloquine was found to interfere with mycolic acids biosynthesis and showed activity against other nontuberculous mycobacteria. Repurposing the well-known antimalarial agent mefloquine could help reduce the time and cost of discovering novel drug candidates.

Genomic characterization of sporadic isolates of the dominant clone of Mycobacterium abscessus subspecies massiliense

Rebecca M. Davidson et al.

Summary: A recent study found a dominant clone of Mycobacterium abscessus subspecies massiliense associated with cystic fibrosis patients, pulmonary outbreaks, and skin infections, with high prevalence in the US and UK. The genomic variation and antimicrobial resistance mutations were compared between different isolates, revealing that outbreak isolates were more likely to have antimicrobial resistance mutations compared to sporadic isolates. Further studies are needed to understand the potential routes of transmission and spread of this clone among non-CF and CF patients in the US.
