Induction of Paraptotic Cell Death in Breast Cancer Cells by a Novel Pyrazolo[3,4-h]quinoline Derivative through ROS Production and Endoplasmic Reticulum Stress

Synergistic killing effect of paclitaxel and honokiol in non-small cell lung cancer cells through paraptosis induction

Summary: The study found that the combination of paclitaxel and honokiol synergistically killed NSCLC cells by inducing paraptosis, characterized by cytoplasmic vacuolation. Mechanistically, proteasomal inhibition-mediated endoplasmic reticulum stress and unfolded protein responses leading to ER dilation, as well as disruption of intracellular Ca(2+) homeostasis and mitochondrial Ca(2+) overload resulting in mitochondrial dysfunction, were involved in paclitaxel/honokiol-induced paraptosis. Additionally, cellular protein protein light chain 3 (LC3) may play a significant role in paclitaxel/honokiol induced cytoplasmic vacuolation and NSCLC cell death. Therefore, the combination of honokiol and paclitaxel may present a novel strategy for the treatment of paclitaxel-resistant NSCLC.

These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
