Summary: Proximal single-stranded RNA significantly impairs the stability of RG4, resulting in easier accessibility of the G-rich sequence at physiological K+ concentrations. This destabilizing effect requires a minimum of six nucleotides of ssRNA and is effective at either end of RG4. Under the influence of such proximal ssRNA, the RG4-forming sequence exhibits interconversions between at least three less stable RG4 conformers.

Folding and persistence times of intramolecular G-quadruplexes transiently embedded in a DNA duplex

Summary: G-quadruplex (G4) DNA structures play important roles in DNA metabolism, and their formation in double-stranded DNA regions is challenged by the complementary strand. By designing a single-molecule assay, the folding and persistence times of G4s in the presence of the complementary strand were measured. It was found that G4s are more stable in tested replication origin and promoters, and the presence of G4 ligands increased both folding rate and persistence time.

Nanomechanics of G-quadruplexes within the promoter of the KIT oncogene
