Epigenetic changes in human model KMT2A leukemias highlight early events during leukemogenesis

Thomas Milan et al.

Summary: Chromosomal translocations involving the KMT2A gene are common in pediatric acute myeloid leukemias. This study investigated the molecular mechanisms of the disease using a human model system. It found that changes in DNA methylation, histone modifications, and chromatin accessibility, as well as expression changes, are associated with leukemic transformation. The study also identified a potential essential gene and signaling pathway involved in leukemogenesis.

JMJD1C knockdown affects myeloid cell lines proliferation, viability, and gemcitabine/carboplatin-sensitivity

Vanessa Schimek et al.

Summary: The study found that JMJD1C is essential for proliferation, survival, and viability of K562 and MEG-01 cells. Additionally, JMJD1C potentially affects the cells' sensitivity to gemcitabine/carboplatin, reducing the IC50 value for both cell lines. Further research is needed to confirm JMJD1C's role in gemcitabine/carboplatin sensitivity, but the findings suggest its potential importance in this area.

Modulators of histone demethylase JMJD1C selectively target leukemic stem cells

Yong Yang et al.

Summary: Leukemic stem cells (LSCs) are a rare cell population that can lead to the development of acute myeloid leukemia. Inhibitors of the histone lysine demethylase JMJD1C, such as JDM-7 and tadalafil, have been identified as potential modulators that selectively inhibit the growth of LSCs.

Comparative Analysis of the Genetic Variants in Haematopoietic Stem/Progenitor and Mesenchymal Stem Cell Compartments in de novo Myelodysplastic Syndromes

W. M. Manoj S. Bandara et al.

Summary: The study identified distinct genetic variations in bone marrow mesenchymal stem cells (MSCs) and CD34(+) HSPCs, with majority being novel potential pathogenic variants. It revealed that DNA methylation, chromatin remodeling, and RNA splicing processes were affected in both cell types, while gene variants in the cohesion complex and epigenetic mechanisms co-existed. These findings suggest that genetic variations occur following lineage commitment in MDS.

Integrative study of EZH2 mutational status, copy number, protein expression and H3K27 trimethylation in AML/MDS patients

Julia Stomper et al.

Summary: Mutations in the EZH2 gene are commonly found in patients with myeloid neoplasms, often co-occurring with other mutations, and are associated with poor prognosis. These mutations result in decreased EZH2 expression and may predict inferior survival outcomes in patients with acute myeloid leukemia receiving standard chemotherapy.

miR-181d/RBP2/NF-κB p65 Feedback Regulation Promotes Chronic Myeloid Leukemia Blast Crisis

Minran Zhou et al.

Summary: Overall, the study found that miR-181d overexpression in CML-BP promotes leukemia cell proliferation. RBP2 was identified as a direct target of miR-181d, inhibiting RBP2 expression and subsequently affecting NF-kappa B p65 transcriptional expression. This feedback regulation contributes to sustained NF-kappa B activation in the development of CML-BP.

Summary: In this study, a new machine learning model ST-123 for predicting AML survival was created using gene expression data. Age, KDM5B, and LAPTM4B were identified as key variables in the model, with high accuracy in predicting survival in high-risk patients and significant prognostic improvements in patients with high-risk mutations.

A KDM4A-PAF1-mediated epigenomic network is essential for acute myeloid leukemia cell self-renewal and survival

Matthew E. Massett et al.

Summary: This study highlights the essential role of KDM4A in acute myeloid leukemia, showing that its loss can induce apoptosis in AML cells. KDM4A controls the epigenomic network essential for AML self-renewal and survival, suggesting its potential as a therapeutic target.

KDM4C contributes to cytarabine resistance in acute myeloid leukemia via regulating the miR-328-3p/CCND2 axis through MALAT1

Lu Xue et al.

Summary: This study demonstrated that KDM4C may upregulate MALAT1 expression, leading to decreased miR-328-3p expression, and subsequently increased CCND2 levels, inducing Ara-C resistance in AML. The findings were further confirmed in an in vivo xenograft NOD/SCID mouse model.

KDM6A promotes imatinib resistance through YY1-mediated transcriptional upregulation of TRKA independently of its demethylase activity in chronic myelogenous leukemia

Summary: The study revealed that abnormal expression of KDM6A in CML leads to resistance to imatinib. By interacting with the NTRK1 gene, KDM6A can activate downstream survival pathways, thereby inducing resistance to imatinib.
