DNA in extracellular vesicles: biological and clinical aspects

Julia Elzanowska et al.

Summary: Research on DNA in extracellular vesicles (EVs) has rapidly evolved in recent years, especially in the field of liquid biopsy, but most studies have focused on RNA or protein content. This review provides a comprehensive discussion on the biology of EV-DNA, including biogenesis, loading mechanisms, and its significant roles in cellular homeostasis, communication, immune response modulation, as well as in clinical settings such as cancer, infections, pregnancy, and prenatal diagnosis.

Summary: This passage discusses the importance of both selective and nonselective mechanisms for RNA release into the extracellular environment, providing new perspectives on RNA-mediated intercellular communication with an analogy to communication through social media. It also highlights how technical limitations have restricted the study of some of the most abundant exRNAs, which may have a stronger impact on recipient cells compared to low abundance miRNAs.

Summary: Extracellular vesicles (EVs) are small vesicles produced by healthy tissues and tumor cells, carrying a rich content of biomolecules, which play important roles in cancer control and progression. They are being studied as carriers of liquid biopsy biomarkers, with double-stranded DNA (dsDNA) in EVs showing potential as a promising biomarker for tumor presence.

Small RNAs are modified with N-glycans and displayed on the surface of living cells

Ryan A. Flynn et al.

Summary: This study challenges the conventional view that RNA is not a major target of glycosylation by providing evidence that mammals use RNA as a third scaffold for glycosylation. The researchers found that conserved small noncoding RNAs bear sialylated glycans, which are present in multiple cell types and mammalian species. The majority of glycoRNAs were located on the cell surface and were shown to interact with anti-dsRNA antibodies and members of the Siglec receptor family, suggesting a direct interface between RNA biology and glycobiology.

Extracellular Vesicles in Synovial Fluid from Rheumatoid Arthritis Patients Contain miRNAs with Capacity to Modulate Inflammation

Andrew D. Foers et al.

Summary: In rheumatoid arthritis, extracellular vesicles are associated with joint inflammation and destruction. Through sequencing EV miRNAs in synovial fluid from RA patients, differences between joints with high- and low-grade inflammation were identified, suggesting a role in regulating inflammation-related genes. EV miRNAs have the potential to modulate inflammation and contribute to RA pathophysiology.

Urinary Extracellular Vesicles and Their miRNA Cargo in Patients with Fabry Nephropathy

Tina Levstek et al.

Summary: Current biomarkers for Fabry nephropathy lack sensitivity in detecting early kidney damage and do not predict progression of nephropathy. However, analysis of urinary extracellular vesicles (uEVs) and their derived microRNAs (miRNAs) may be applicable in predicting the development and progression of nephropathy in Fabry disease. The total concentration and size of uEVs were not indicative of nephropathy presence or progression, while certain miRNAs may play a role in Fabry nephropathy but further studies are needed to confirm their significance.
