Mechanical Feed-Forward Loops Contribute to Idiopathic Pulmonary Fibrosis

Margaret A. T. Freeberg et al.

Summary: Idiopathic pulmonary fibrosis is a progressive disease characterized by accumulation of extracellular matrix in lung tissue, leading to altered mechanical properties. Recent studies suggest that these changes create a feed-forward loop, contributing to fibroblast differentiation and activation. Mechanotransduction pathways, including mechanosensors like integrins and downstream transcription factors, play a role in fibrosis progression.

The role of activin: the other side of chronic kidney disease-mineral bone disorder?

Giuseppe Cianciolo et al.

Summary: Chronic kidney disease-mineral bone disorder (CKD-MBD) is closely linked to elevated cardiovascular morbidity and mortality in CKD patients. Emerging evidence suggests that pathways like canonical Wnt signaling are activated early in CKD and may play a role in the development of adynamic bone disease and vascular calcification. Studies show that the progression of vascular calcification parallels changes in bone mineral density and turnover, indicating shared pathways between bone and arterial mineralization. Further research is needed to identify new mediators involved in CKD-MBD pathogenesis and develop more effective therapeutic strategies.

Bioinspired in vitro microenvironments to control cell fate: focus on macromolecular crowding

Dimitrios I. Zeugolis

Summary: The development of therapeutic regenerative medicine and accurate drug discovery cell-based products requires effective cell expansion ex vivo. Traditional cell culture systems often fail to recapitulate tissue environments, leading to changes in cell phenotype and function, prompting the need for new approaches to maintain cell characteristics.

Intraductal xenografts show lobular carcinoma cells rely on their own extracellular matrix and LOXL1

George Sflomos et al.

Summary: Invasive lobular carcinoma (ILC) is a common subtype of breast cancer characterized by loss of E-cadherin. This study used mouse models to recapitulate ILC, identifying extracellular matrix modulation as a intrinsic trait of lobular carcinoma cells. Inhibition of LOXL1 shows potential as a therapeutic strategy for ILC by disrupting ECM structure and decreasing tumor growth, invasion, and metastasis.

Transforming eukaryotic cell culture with macromolecular crowding

Michael Raghunath et al.

Summary: Standard eukaryotic cell culture lacks the density and complexity of macromolecular species present in tissues, making it difficult for cells to perform physiologically. The use of macromolecular crowding (MMC) in eukaryotic cell culture is proposed for regenerative medicine and drug discovery purposes to better mimic the crowded environment of tissues.
