m6A-Atlas: a comprehensive knowledgebase for unraveling the N6-methyladenosine (m6A) epitranscriptome

Yujiao Tang et al.

Summary: N-6-Methyladenosine (m(6)A) is the most prevalent RNA modification on mRNAs and lncRNAs, and m(6)A-Atlas is a comprehensive knowledgebase for unraveling the m(6)A epitranscriptome. It features a high-confidence collection of m(6)A sites and quantitative epitranscriptome profiles, offering novel features like conservation among vertebrate species and potential pathogenesis inference from disease-associated genetic mutations. The user-friendly interface allows for querying, visualization, and sharing of annotated m(6)A epitranscriptomes.

RMVar: an updated database of functional variants involved in RNA modifications

Xiaotong Luo et al.

Summary: Distinguishing disease-related variants within a large number of variants is challenging. The updated RMVar database includes 1,678,126 variants associated with 9 RNA modifications, offering a new perspective on the effects of RM-associated variants on posttranscriptional regulation, as well as integrating disease-related information for further functional studies on genetic variants affecting RNA modifications.

Exome sequencing and analysis of 454,787 UK Biobank participants

Joshua D. Backman et al.

Summary: In this study, whole-exome sequencing was used to identify gene-trait associations in 454,787 individuals, revealing 564 distinct genes with significant trait associations. Rare variant associations were enriched in loci from genome-wide association studies (GWAS) but most were independent of common variant signals.

Genome-wide enhancer maps link risk variants to disease genes

Joseph Nasser et al.

Summary: The study utilized the ABC model to create enhancer-gene maps in 131 human cell types and tissues and interpreted the functions of GWAS variants in 72 diseases and complex traits. It was found that in inflammatory bowel disease, causal variants are enriched in predicted enhancers in specific cell types, and the ABC model shows higher precision in connecting noncoding variants to target genes.

m6A demethylase ALKBH5 controls CD4+ T cell pathogenicity and promotes autoimmunity

Jing Zhou et al.

Summary: The m(6)A eraser ALKBH5 plays a crucial role in controlling the pathogenicity of CD4(+) T cells during autoimmunity, as its deficiency results in weakened immune responses and reduced recruitment of neutrophils into the central nervous system.
