CD44 engagement enhances acute myeloid leukemia cell adhesion to the bone marrow microenvironment by increasing VLA-4 avidity

Julia C. Gutjahr et al.

Summary: The cooperation of CD44 and VLA-4 in AML was found to enhance leukemia cell adhesion on VCAM-1, leading to activation of Akt, MAPK, NF-kappa B and mTOR signaling pathways and decreased cell apoptosis. This mechanism provides insights into the regulation of AML cell retention in the bone marrow microenvironment.

CD79a promotes CNS-infiltration and leukemia engraftment in pediatric B-cell precursor acute lymphoblastic leukemia

Lennart Lenk et al.

Summary: Lenk et al identify CD79a as a key component associated with CNS infiltration and relapse in B-cell precursor (BCP)-ALL patients. They demonstrate that downregulation of CD79a inhibits the engraftment of leukemia cells in various murine xenograft models, particularly in the CNS.

Reactive Oxygen Species Rewires Metabolic Activity in Acute Myeloid Leukemia

Andrew J. Robinson et al.

Summary: In AML patients, over 60% exhibit constitutive activation of NOX2 and increased ROS production. This leads to increased glucose uptake and proliferation in AML cells, as well as changes in carbohydrate metabolism. Metabolomic profiling revealed significant ROS-specific metabolic alterations in sphingolipid metabolism, fatty acid oxidation, purine metabolism, amino acid homeostasis, and glycolysis, providing further evidence of ROS rewiring metabolism in AML.

RUNX2 regulates leukemic cell metabolism and chemotaxis in high-risk T cell acute lymphoblastic leukemia

Summary: The study demonstrates that the upregulation of RUNX2 acts as a dependency factor in high-risk subtypes of human T-ALL by regulating tumor metabolism and leukemic cell migration simultaneously.

MAPK-ERK is a central pathway in T-cell acute lymphoblastic leukemia that drives steroid resistance

Jordy C. G. van der Zwet et al.

Summary: Activation of IL7R signaling contributes to steroid resistance in pediatric T-ALL patients through phosphorylation of BIM, which can be restored by MEK inhibitors. Selumetinib shows synergistic effects with steroids in both IL7-dependent and IL7-independent steroid resistant T-ALL PDX samples, highlighting its potential as a treatment option.

Adipocytes Provide Fatty Acids to Acute Lymphoblastic Leukemia Cells

Jonathan Tucci et al.

Summary: The study found that adipocytes release free fatty acids in the presence of acute lymphoblastic leukemia cells, which are taken up by the leukemia cells and incorporated into triglycerides and phospholipids. Adipocyte-derived lipids can relieve ALL cell endogenous lipogenesis and reverse the cytotoxicity of chemotherapy, altering cell metabolism from glucose to fatty acid oxidation. Targeting lipid synthesis and metabolism may potentially reverse adipocyte protection of ALL cells.

Insights on Metabolic Reprogramming and Its Therapeutic Potential in Acute Leukemia

Ludovica Di Martino et al.

Summary: Acute leukemias are the most common hematologic tumors in adolescents and young adults, with metabolic reprogramming playing a crucial role in their pathogenesis and treatment. This review summarizes the main metabolic pathways contributing to the growth, redox balance, and drug resistance of leukemia cells, identifying vulnerabilities that could be targeted for therapy.

Stabilization of fatty acid synthesis enzyme acetyl-CoA carboxylase 1 suppresses acute myeloid leukemia development

Hidenori Ito et al.

Summary: The study demonstrates that ACC1 plays a critical role in regulating the growth and differentiation of leukemia-initiating cells, with its stable expression inhibiting AML development and promoting terminal differentiation of Trib1-COP1-expressing cells to eradicate leukemia-initiating cells. ACC1 upregulation shows potential as an effective strategy for cancer therapy.

Leukaemia: a model metastatic disease

Andrew E. Whiteley et al.

Summary: Leukaemia cells possess innate migration and invasion abilities, unlike solid tumours which require genetic modifications for effective metastasis. Therefore, although traditionally not considered as metastatic diseases, leukaemias are actually models of highly efficient metastatic spread and are challenging to treat.
