Nicotine promotes vascular calcification via intracellular Ca2+-mediated, Nox5-induced oxidative stress, and extracellular vesicle release in vascular smooth muscle cells

Summary: This study investigates the effects of nicotine on vascular smooth muscle cell calcification and its underlying mechanisms. The results show that calcification is more common in carotid plaques of smokers, and nicotine induces calcification and increases the expression of calcification-related genes and extracellular vesicle secretion in vascular smooth muscle cells. The pro-calcifying effects of nicotine are mediated by Ca2+-dependent Nox5, and Nox5 expression is higher in carotid arteries of smokers.

Treatment with curcumin and nicotinic-curcumin can reduce endothelial EVs secretion, potentially by inhibiting inflammation.

Poor prognosis of PAD may lead to impaired walking function and increased risk of cardiovascular events. Reliable biomarkers and machine learning methods have the potential to improve identification and personalized treatment of PAD patients.

Role of Runx2 in Calcific Aortic Valve Disease in Mouse Models

Summary: In this study, it was found that aVICs and sinus wall cells give rise to Runx2+ osteochondrogenic cells in diseased mouse aortic valves. Conditional depletion of Runx2 in specific cells led to decreased osteochondrogenic gene expression and improved aortic valve function in a mouse model of CAVD with high prevalence of hemodynamic valve dysfunction. Leaflet hinge and sinus wall calcification were significantly decreased following Runx2 depletion, suggesting a potential causal role in valve dysfunction.
