18S rRNA gene sequence-structure phylogeny of the Trypanosomatida (Kinetoplastea, Euglenozoa) with special reference to Trypanosoma

Alyssa R. Borges et al.

Summary: In this study, the secondary structures of 18S rRNA gene from Trypanosoma cruzi and Trypanosoma brucei were predicted by homology modeling. Sequence-structure trees were generated by aligning sequences with their secondary structures, providing insights into the phylogeny of Trypanosomatida and a comprehensive sampling of trypanosomes. The robust support for well-known clades and critical branches indicates the potential of using 18S rRNA sequence and secondary structure data for reconstructing phylogenetic trees in trypanosomatids.

Structure of trypanosome coat protein VSGsur and function in suramin resistance

Johan Zeelen et al.

Summary: Recent studies have shown that trypanosome strains expressing VSGsur have higher resistance to suramin. The tightly bound VSGsur and suramin defines a structurally divergent subgroup of coat proteins, with the co-crystal structure revealing how the drug binds asymmetrically within the VSG homodimer. Loss-of-contact mutations in VSGsur significantly reduce affinity to suramin, indicating that the resistance phenotype is dependent on the binding of suramin to VSGsur.

An invariant Trypanosoma vivax vaccine antigen induces protective immunity

Delphine Autheman et al.

Summary: A systematic genome-led approach identified protective invariant subunit vaccine antigens for Trypanosoma vivax, providing a vaccine candidate for the important parasitic disease that has constrained socioeconomic development in sub-Saharan Africa. The study showed that highly protective vaccines against trypanosome infections can be achieved, despite the parasite's sophisticated immunoprotective mechanisms.

Dynamic, variable oligomerization and the trafficking of variant surface glycoproteins of Trypanosoma brucei

To the Surface and Back: Exo- and Endocytic Pathways in Trypanosoma brucei

Fabian Link et al.

Summary: Trypanosoma brucei is a unicellular pathogen that thrives extracellularly in vertebrate hosts, with its cell surface playing a critical role in both immune recognition and evasion. The variant surface glycoprotein (VSG) and incessant motility contribute to immune evasion, while endo- and exocytosis occur at the flagellar pocket. Mechanisms behind VSG recycling and secretion are not fully understood, presenting longstanding questions that may be answered with novel technologies in the future.
