Pathobiological Origins and Evolutionary History of Highly Pathogenic Avian Influenza Viruses

Summary: Over the past 60 years, 42 natural, independent H5 and H7 low-pathogenic avian influenza virus conversion events to highly pathogenic viruses have occurred in Europe, North America, Oceania, Asia, Africa, and South America. While most outbreaks were limited and eradicated, some outbreaks caused by H5Nx, H7N3, and H7N9 viruses continue to persist. Active surveillance and molecular detection efforts are crucial for early detection and eradication of these viruses in domestic birds.

Summary: Multiple basic amino acids at the protease cleavage site of HA protein play a key role in the virulence of highly pathogenic avian influenza viruses. Recombination of HA RNA with host RNAs, particularly at hotspots in rRNAs, tRNAs, and viral RNAs, is a dominant mechanism for acquiring MBCS in H7 subtype HPAI. The presence of small nucleolar RNAs near recombination points facilitates RNA recombination in highly abundant RNAs.
