A long-half-life, high-affinity P-selectin inhibitor

Summary: P-G6, a pegylated glycomimetic of the N terminus of P-selectin glycoprotein ligand (PSGL-1), inhibits the P-selectin-PSGL-1 interaction and shows potential for prevention and treatment of venous thromboembolism (VTE). The pegylation endows P-G6 with a longer half-life and it has shown effectiveness in inhibiting thrombosis in preclinical studies without prolonging bleeding time.

Peptidylarginine Deiminase Inhibition Prevents Diabetes Development in NOD Mice

Summary: In this study, systemic inhibition of peptidylarginine deiminases (PADs) through BB-Cl-amidine treatment prevented diabetes development in NOD mice by reducing citrullination levels in the pancreas, decreasing circulating autoantibody titers, and reducing neutrophil extracellular trap formation. Additionally, the treatment induced a shift in cytokine production and increased regulatory T cell frequency. This highlights the potential of PAD inhibition for treating autoimmune diseases like type 1 diabetes.

Summary: Type 1 diabetes is characterized by insulin deficiency due to selective destruction of pancreatic beta-cells by self-reactive T cells. Recent evidence suggests that innate immune responses play a significant role in the pathogenesis of T1D, with the accumulation of leukocytes in the pancreas contributing to the chronic progression of pancreatic damage. Genetic and environmental factors have differing impacts on the etiology of T1D, which is specific to each patient.

Neutrophil-Associated Inflammatory Changes in the Pre-Diabetic Pancreas of Early-Age NOD Mice

Summary: The accumulation of neutrophils and neutrophil elastase in the pancreas of NOD mice as early as 2 weeks of age indicates inflammatory changes that precede the development of diabetes. Inhibiting key neutrophil enzymes did not prevent the progression to hyperglycemia in the NOD mice, suggesting that other effector cells may also play a role in the development of autoimmune diabetes.

Type 1 diabetes mellitus: much progress, many opportunities

Although there is considerable heterogeneity in genetics, pathology, and clinical course of T1D, there are still knowledge gaps and opportunities for improving understanding of its pathogenesis. Emerging therapies are being developed to treat or prevent T1D.
