Influenza infection fortifies local lymph nodes to promote lung-resident heterosubtypic immunity

Daniel H. Paik et al.

Summary: This study demonstrates that during influenza infection, lung TRM cells are generated and can mediate protective immunity, with lung T cell responses derived from both TRMs and effector T cells generated in the local lung draining mediastinal lymph nodes. The initial infection strengthens the medLNs with an increased number of cDCs, enhancing the priming of T cells, including those specific for new epitopes. Depletion of cDCs during the recall response reduces medLN T cell generation and heterosubtypic immunity.

CCR2-and Flt3-Dependent Inflammatory Conventional Type 2 Dendritic Cells Are Necessary for the Induction of Adaptive Immunity by the Human Vaccine Adjuvant System AS01

Cedric Bosteels et al.

Summary: The AS01 adjuvant system containing MPL and QS-21 in liposomal formulation is crucial for the recruitment and activation of dendritic cells, particularly cDC2s and inf-cDC2s, which play a significant role in priming antigen-specific T cell responses post-vaccination. Depletion of these DC subsets results in the abrogation of adaptive immune responses, highlighting their importance in the adjuvant activity of AS01.
