Summary: This study revealed that the loss of XIAP gene associated with Mendelian IBD rendered Paneth cells sensitive to microbiota and factors like TNF, RIPK1, and RIPK3, leading to deficiency in antimicrobial peptides and alterations in microbiota composition. While loss of XIAP alone did not cause intestinal inflammation, it did make the host susceptible to pathobionts that can promote granulomatous ileitis.
