Whole-body senescent cell clearance alleviates age-related brain inflammation and cognitive impairment in mice

Mikolaj Ogrodnik et al.

Summary: Clearing senescent cells, either through drug treatment or senolytic cocktail, has been shown to improve cognitive function, providing proof-of-concept for senolytic interventions as a potential therapeutic avenue for alleviating age-associated cognitive impairment.

Neuroblast senescence in the aged brain augments natural kill cell cytotoxicity leading to impaired neurogenesis and cognition

Summary: The accumulation of natural killer (NK) cells in the aging brain impairs neurogenesis and cognition, as neuroblast senescence in the dentate gyrus augments NK cell cytotoxicity during normal brain aging.

Synaptic elimination by microglia and disturbed higher brain functions

Kazuya Miyanishi et al.

Summary: Microglial cells in normal mature brains play a crucial role in phagocytosing synapses to maintain neural circuits and affect brain functions. Impaired synaptic elimination by microglia can disrupt the E/I balance, leading to various problems in memory, cognition, sleep, movement, social behaviors, and thinking. Additionally, disturbances in microglial phagocytosis functions are linked to neurological, mental, and developmental disorders like Parkinson's and Alzheimer's diseases, autism spectrum disorder, ADHD, and schizophrenia.

Demonstration of age-related blood-brain barrier disruption and cerebromicrovascular rarefaction in mice by longitudinal intravital two-photon microscopy and optical coherence tomography

Adam Nyul-Toth et al.

Summary: Age-related blood-brain barrier disruption and cerebromicrovascular rarefaction play important roles in the pathogenesis of vascular cognitive impairment and dementia (VCID) and Alzheimer's disease (AD). Recent advances in geroscience research have provided new interventions for preventing these diseases. Sensitive and easy-to-adapt imaging methods are urgently needed for longitudinal assessment of changes in BBB permeability and brain capillarization in aged mice.

White matter aging drives microglial diversity

Shima Safaiyan et al.

Summary: Research has identified white matter-associated microglia (WAMs) as a protective response during aging, characterized by phagocytic activity and lipid metabolism gene activation. WAMs depend on triggering receptor expressed on myeloid cells 2 (TREM2) signaling and are independent of apolipoprotein E (APOE) in contrast to mouse models of Alzheimer's disease.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.

Treatment with the BCL-2/BCL-xL inhibitor senolytic drug ABT263/Navitoclax improves functional hyperemia in aged mice

Stefano Tarantini et al.

Summary: The present study found that NVC responses were significantly impaired in aged mice, and treatment with ABT263/Navitoclax improved NVC response and hippocampal-encoded functions of learning and memory. The findings suggest that senescent cells in the aged brain may contribute to age-related neurovascular uncoupling, exacerbating cognitive decline. The neurovascular protective effects of ABT263/Navitoclax treatment highlight the potential of senolytic treatments as effective interventions in patients at risk for vascular cognitive impairment.
