Summary: The research focused on developing and optimizing nanoniosomes for delivering glibenclamide as a hypoglycemic agent to the lungs through an inhaler dosage form. Fifteen different niosomal dispersion formulations were prepared, and the impact of drug amount, cholesterol molar ratio, and HLB values of the surfactant on various characteristics of the nanoniosomes was studied. The optimized formula showed promising results in terms of vesicle size, PDI, ZP, and in-vitro drug release, with in-vivo studies demonstrating efficient delivery and significant hypoglycemic effects in hyperglycemic rats.
