Chylomicrons-Simulating Sustained Drug Release in Mesenteric Lymphatics for the Treatment of Crohn's-Like Colitis

Yi Yin et al.

Summary: The study developed a nano-delivery system to enhance drug delivery in mesenteric lymphatic tissue and evaluated its therapeutic effects in Crohn's colitis. The results showed that the system effectively increased drug concentration in MLT, suppressed lymphangitis, and promoted lymphatic drainage in experimental colitis models. The system also downregulated pro-inflammatory cytokines, inhibited lymphangiogenesis, and ameliorated intestinal inflammation compared to free drug administration.

Targeted delivery of mycophenolic acid to the mesenteric lymph node using a triglyceride mimetic prodrug approach enhances gut-specific immunomodulation in mice

Ruby Kochappan et al.

Summary: The mesenteric lymph nodes play a crucial role in generating adaptive immune responses and contribute to the pathophysiology of various conditions; targeting immunomodulating drugs to the MLN may be beneficial. Using a triglyceride mimetic prodrug approach, this study found that delivering mycophenolic acid in prodrug form resulted in increased lymphatic transport and MLN concentrations, demonstrating potential pharmacodynamic benefits of targeting the MLN with this technology.

Identification, isolation and analysis of human gut-associated lymphoid tissues

Peter B. Jorgensen et al.

Summary: The authors present a protocol for isolating and studying human gut-associated lymphoid tissue, which allows identification and isolation of all human GALT and GALT-free intestinal lamina propria. This technique involves mechanical separation of intestinal mucosa from the submucosa to identify and isolate specific types of lymphoid tissue for downstream cellular and molecular characterization. The protocol, taking 4-10 hours, could be useful for researchers interested in intestinal immune development and function in health and disease.

Transmucosal Solid Lipid Nanoparticles to Improve Genistein Absorption via Intestinal Lymphatic Transport

Antonella Obinu et al.

Summary: In this study, solid lipid nanoparticles (SLN) loaded with GEN were developed to improve its oral bioavailability by avoiding first-pass metabolism through intestinal lymphatic absorption. The SLN obtained by hot homogenization showed high stability and drug loading capacity.

Human gut-associated lymphoid tissues (GALT); diversity, structure, and function

Urs M. Morbe et al.

Summary: Human GALT, including Peyer's patches, appendix, and ILF, plays a key role in antigen sampling and adaptive immune responses in the intestinal wall. Current understanding of human GALT is primarily based on studies in mice, and its relevance to human health remains unclear. Further research is needed to explore the structure, composition, and potential regulatory functions of human GALT in intestinal immune responses.

Summary: The study demonstrates successful in vivo gene silencing in a selective subset of leukocytes using a conformation-sensitive targeting strategy, showing potential therapeutic applications in a murine model of colitis. The results suggest that this lipid nanoparticle targeting strategy could be applied for selective delivery of payloads to other conformation-sensitive targets.

Nano and Microparticles as Potential Oral Vaccine Carriers and Adjuvants Against Infectious Diseases

Seyed Davoud Jazayeri et al.

Summary: Mucosal surfaces are the initial site of infection for most diseases, making oral vaccination crucial for providing protection as the first line of defense through stimulation of immune responses at both systemic and mucosal levels. Successful oral vaccines must overcome the challenges posed by the harsh gastrointestinal environment and low immunogenicity, which can be addressed through the development of delivery systems and adjuvants like nanoparticles and microparticles.
