Confirmation of plant-derived exosomes as bioactive substances for skin application through comparative analysis of keratinocyte transcriptome

Jeong Hun Cho et al.

Summary: Plant exosomes, as nanosized membrane vesicles, have been found to differ in efficacy from conventional plant extracts. They have a stronger impact on the transcriptome and gene expression related to aging, regeneration, skin barrier, and moisturization. Additionally, different plant exosomes show specific effects depending on the plant they are derived from. These findings suggest that plant exosomes have the potential to be commercialized as cosmeceutical products.

Effective methods for isolation and purification of extracellular vesicles from plants

Yifan Huang et al.

Summary: This study compares various methods for isolating plant extracellular vesicles (EVs) and recommends a detailed protocol involving differential centrifugation, density gradient ultracentrifugation, and immunoaffinity capture for isolating and purifying specific subclasses of EVs. Immunoaffinity capture is found to be the most precise method for specific EV subclass isolation when suitable specific EV biomarkers and corresponding antibodies are available. Overall, the study serves as a guide for selecting and optimizing EV isolation methods for desired downstream applications.

Lyophilization Preserves the Intrinsic Cardioprotective Activity of Bioinspired Cell-Derived Nanovesicles

Yub Raj Neupane et al.

Summary: The study introduced a lyophilization approach to preserve critical characteristics of cell-derived nanovesicles (CDNs) for long-term storage, ensuring structural integrity and biological activity. Trehalose as a lyoprotectant raised the glass transition temperature and reduced residual moisture content of CDNs, maintaining the secondary structure of cellular proteins. Both lyoprotected and freshly prepared CDNs targeted and protected injured hearts within 24 hours in a myocardial ischemia/reperfusion injury animal model, highlighting their potential as stable cell-based therapeutics.

Cyclic tangential flow filtration system for isolation of extracellular vesicles

Kimin Kim et al.

Summary: The study introduces a cyclic tangential flow filtration system that can better isolate EVs in the 30-200nm size range through dual cyclic filtration. By introducing a buffer-exchange process after filtration, the purification efficiency is improved. EVs isolated by the cyclic TFF system exhibit more abundant exosome markers and active EVs compared to conventional methods.
