Safety, Pharmacokinetics, and Causal Prophylactic Efficacy of KAF156 in a Plasmodium falciparum Human Infection Study

James G. Kublin et al.

Summary: KAF156, a novel antimalarial drug with high protective efficacy, was found to be safe and well tolerated in a controlled human malaria infection model. Different doses of KAF156 showed varying levels of protection against malaria infection, with higher doses providing greater protection.

Advances in nanomaterial vaccine strategies to address infectious diseases impacting global health

Chelsea N. Fries et al.

Summary: Nanotechnology is being utilized to design new vaccines and facilitate their global implementation, particularly for globally devastating diseases such as HIV, malaria, and tuberculosis. Successful vaccines will need to trigger immune responses different from those raised by normal infection, and nanomaterials provide strategies for achieving this.

Triple Artemisinin-Based Combination Therapies for Malaria - A New Paradigm?

Rob W. van der Pluijm et al.

Summary: Recent progress in the fight against malaria is at risk due to the emergence and spread of artemisinin and partner drug resistance, particularly in the Greater Mekong Subregion. New drugs are not expected in the near future, making it crucial to utilize triple artemisinin-based combination therapies to combat falciparum malaria effectively. Deployment of such therapies beyond the GMS could potentially help prevent global resistance to currently available drugs.

Zwitterionic self-assembled nanoparticles as carriers for Plasmodium targeting in malaria oral treatment

Arnau Biosca et al.

Summary: Novel nanoparticles were designed and synthesized to enhance the bioavailability of antimalarials, showing specific targeting to Plasmodium falciparum-infected red blood cells with maintained in vitro efficacy upon encapsulation of the lipophilic antimalarial drug curcumin. In vivo assays demonstrated increased curcumin concentration in mouse blood after oral administration of the nanoparticles, leading to similar antimalarial activities compared to free drug in Plasmodium yoelii yoelii-infected mice. Microscopic analysis revealed the presence of the polymer inside parasitized erythrocytes one hour after oral administration to infected animals.

Mohamed Farouk Chughlay et al.

Summary: P218 demonstrated excellent chemoprotective efficacy against P. falciparum, effectively inhibiting the parasite. Clinical trials showed that P218 had good safety and tolerability in participants, with mild, transient, and self-limiting adverse events. Furthermore, P218's pharmacokinetics were similar in participants with and without induced infection.
