An organoid-based drug screening identified a menin-MLL inhibitor for endometrial cancer through regulating the HIF pathway

Jingyao Chen et al.

Summary: Tumor organoids were used for drug screening and MI-136 was identified as a potential inhibitor for endometrial cancer by targeting the HIF pathway. The upregulation of MEN1 in human endometrial cancers was found to be closely correlated with the expression levels of HIF1A.

Current status and future outlook for patient-derived cancer models from a rare cancer research perspective

Tadashi Kondo

Summary: Rare cancers, with low incidences but diverse genotypes and phenotypes, pose challenges in designing clinical trials due to limited patient numbers, resulting in poor clinical outcomes and lack of effective anticancer drugs. Adequate cancer models faithfully recapitulating tumor features are crucial for focusing on more efficacious drugs in clinical studies. Patient-derived cancer models are particularly important for rare cancers, but few are currently available.

Recapitulating macro-scale tissue self-organization through organoid bioprinting

Jonathan A. Brassard et al.

Summary: The 3D bioprinting approach combines organoid-forming stem cells and extracellular matrix to achieve tissue self-organization, allowing for the generation of tissues with complex structures and diverse cell types at centimeter scales. This method opens up new avenues for drug discovery, diagnostics, and regenerative medicine by controlling tissue self-organization from millimeter to centimeter scales.

Toward a neurospheroid niche model: optimizing embedded 3D bioprinting for fabrication of neurospheroid brain-like co-culture constructs

Yi-Chen Ethan Li et al.

Summary: Researchers have used embedded 3D bioprinting to create structured brain-like co-culture constructs with neurospheroid patterns, mimicking a supportive bed resembling a neural stem cell growth environment. These brain-like co-cultures can provide a reproducible platform for modeling neurological diseases, neural regeneration, and drug development and repurposing.

Hedgehog transcriptional effector GLI mediates mTOR-Induced PD-L1 expression in gastric cancer organoids

Vivien Koh et al.

Summary: The study shows that PD-L1 expression in gastric cancer is mediated by Hh and mTOR signaling pathways. Patient-derived organoid/immune cell co-cultures can be used to study immunosuppressive MDSC function. DSP analysis reveals infiltration of immunosuppressive MDSCs in gastric cancer tissues.

Controlling the polarity of human gastrointestinal organoids to investigate epithelial biology and infectious diseases

Julia Y. Co et al.

Summary: The polarity of gastrointestinal organoids is reversed to study epithelial biology and host-microbe interactions, allowing increased access to the apical surface while preserving functions. The method quickly establishes an outward-facing apical surface, maintaining epithelial integrity, secretory and absorptive functions, and enabling differentiation control. This technique enhances the possibilities of organoid use in gastrointestinal and respiratory health and disease research by controlling organoid polarity.
