Clinical relevance of extracellular vesicles in hematological neoplasms: from liquid biopsy to cell biopsy

Summary: Liquid biopsy is becoming increasingly important in oncology within the era of precision medicine, allowing for the isolation and analysis of tumor-derived biomarkers such as extracellular vesicles (EVs) in body fluids. EVs, as lipid bilayer-enclosed particles, have emerged as valuable carriers of cancer information, offering potential as novel biomarkers due to their molecular cargo mirroring parental cell characteristics. The translation of these new diagnostic tools into clinical practice has the potential to deeply revolutionize the field of cancer research.

Distinct profile of CD34+ cells and plasma-derived extracellular vesicles from triple-negative patients with Myelofibrosis reveals potential markers of aggressive disease

Summary: In this study, it was found that the absolute numbers of circulating CD34(+) cells were significantly increased in triple-negative (TN) myelofibrosis patients. These cells displayed in vitro defective functions and were unresponsive to the inflammatory microenvironment. Furthermore, the circulating extracellular vesicles (EVs) from TN patients were found to promote the survival of TN CD34(+) cells, suggesting a distinct EV-driven signal promoting malignant hemopoiesis in TN patients. The distinct gene expression profiling analysis and microRNA signature in TN patients may provide insights for novel therapeutic approaches for this rare form of myelofibrosis.
