Belzutifan: First Approval

Emma D. Deeks

Summary: Belzutifan is an oral small molecule inhibitor of hypoxia-inducible factor (HIF)-2 alpha, developed by Peloton Therapeutics for the treatment of solid tumors, including renal cell carcinoma (RCC) and von Hippel-Lindau (VHL) disease-associated RCC. It has received its first approval in the USA for the treatment of VHL disease patients requiring therapy for associated tumors, with clinical studies ongoing in other indications.

Quantitative Proteomics Identifies Secreted Diagnostic Biomarkers as well as Tumor-Dependent Prognostic Targets for Clear Cell Renal Cell Carcinoma

Aydanur Senturk et al.

Summary: Through in-depth quantitative proteomics analysis, the study identified SPARC as a promising urine biomarker for ccRCC, providing new insights for disease detection; proposed a biomarker panel that classifies ccRCC tumors into two clusters, with cluster 1 tumors having a significantly higher risk of death compared to cluster 2 tumors.

Inhibition of hypoxia-inducible factor-2α in renal cell carcinoma with belzutifan: a phase 1 trial and biomarker analysis

Toni K. Choueiri et al.

Summary: The HIF-2 alpha inhibitor belzutifan shows promising clinical activity and a favorable safety profile in heavily pre-treated patients with renal cell carcinoma, suggesting it may offer an effective treatment option. The drug was well tolerated with a confirmed objective response rate of 25% and a median progression-free survival of 14.5 months in patients with ccRCC.

The role of the extracellular matrix protein TGFBI in cancer

Armando Corona et al.

Summary: TGFBI plays a dual role in cancer progression, acting as a tumor suppressor in the context of hereditary corneal dystrophies and promoting tumor progression in established tumors. Targeting TGFBI may have potential clinical utility in treating advanced cancers and assessing TGFBI levels could serve as a biomarker for chemotherapy resistance and tumor progression.

Clear cell renal cell carcinoma ontogeny and mechanisms of lethality

Eric Jonasch et al.

Summary: The molecular features that define the initiation and progression of clear cell renal cell carcinoma (ccRCC) are increasingly being understood. Common genomic and chromosomal copy number abnormalities in ccRCC can be categorized into initiating events, drivers of progression, and factors that confer lethality. Mutations in specific genes in ccRCC may affect genomic stability and immune cell populations within tumors.
