Summary: Acetic acid fermentation in Gluconacetobacter diazotrophicus strain PAL5 involves the oxidation of ethanol to acetic acid via acetaldehyde, catalyzed by membrane-bound alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH). The study identified that ALDH activity in PAL5 is derived from three enzyme complexes, with AldFGH playing a major role and requiring molybdopterin rather than PQQ for its activity. Deletion of the PQQ biosynthesis gene cluster abolished ADH activity completely but did not affect ALDH activity.
