Preclinical development of a humanized chimeric antigen receptor against B-cell maturation antigen for multiple myeloma

Lorena Perez-Amill et al.

Summary: This study developed CAR-T cells targeting multiple myeloma, with ARI2h cells showing superior therapeutic efficacy compared to ARI2m cells, especially in cases with high tumor burden disease. The humanization of the single chain variable fragment and large-scale expansion of CAR-T cells set the groundwork for a clinical trial to evaluate the efficacy of ARI2h cell treatment for multiple myeloma patients.

miR-21 antagonism abrogates Th17 tumor promoting functions in multiple myeloma

Marco Rossi et al.

Summary: The study demonstrated that miR-21 plays a crucial role in Th17-mediated tumor growth and bone disease in multiple myeloma, with inhibition of miR-21 leading to reduced pathology. Early inhibition of miR-21 in T cells (miR-21i-T cells) was found to impair Th17 differentiation, thereby decreasing MM cell proliferation and osteoclast activity.

Single-cell profiling reveals the trajectories of natural killer cell differentiation in bone marrow and a stress signature induced by acute myeloid leukemia

Adeline Crinier et al.

Summary: NK cells in human bone marrow can be classified into three subpopulations, with NK0 cells believed to be precursors of circulating CD56(dim) NK1-like NK cells and CD56(bright) NK2-like NK cells under physiological conditions. Transcriptomic analysis showed stress-induced repression of NK cell effector functions in bone marrow NK cells from patients with acute myeloid leukemia (AML), with reduced levels of CD160, but higher survival rates in the CD160(high) group.

Chronic inflammation towards cancer incidence: A systematic review and meta-analysis of epidemiological studies

Nathalie Michels et al.

Summary: This study conducts a systematic review and meta-analysis to explore the association between chronic inflammation and cancer incidence. It finds that specific inflammatory markers like CRP, fibrinogen, and IL6 are related to certain cancer types such as breast, colorectal, and lung cancer. Improvements in study quality could be made by verifying inflammatory status, adjusting for confounders, and ensuring long-term follow-up.

Comprehensive Profiling of an Aging Immune System Reveals Clonal GZMK+ CD8+ T Cells as Conserved Hallmark of Inflammaging

Denis A. Mogilenko et al.

Summary: This study characterized age-associated alterations in immune cells across multiple mouse organs and identified a distinct population of age-associated GZMK-expressing CD8(+) T cells. These Taa cells, highly clonal with specific features, were found to be a potential target to address age-associated dysfunctions of the immune system. In humans, proportions of the circulating GZMK(+) CDE3(+) T cell population that shares signatures with mouse Taa cells also increased during healthy aging.

A systematic review and meta-analysis to inform cancer screening guidelines in idiopathic inflammatory myopathies

Alexander G. S. Oldroyd et al.

Summary: Clinical factors associated with increased and decreased cancer risk in IIM patients were identified through systematic literature review and meta-analysis. Factors such as DM subtype, older age, male sex, dysphagia, cutaneous ulceration, and anti-transcriptional intermediary factor were found to increase cancer risk, while factors like PM subtype, Raynaud's phenomenon, interstitial lung disease, high serum creatine kinase or lactate dehydrogenase levels, and certain autoantibody positivity were associated with reduced cancer risk. Additionally, limited evidence suggested that CT scanning could be effective for cancer screening in IIM patients, highlighting the need for further research and consensus guidelines.

Thymic atrophy creates holes in Treg-mediated immuno-regulation via impairment of an antigen-specific clone

Rachel Thomas et al.

Summary: Age-related thymic atrophy reduces the output of naive conventional T cells, but does not affect the generation of pan-Treg cells. Specific tissue-specific Treg clones may experience abnormal agonist selection due to thymic atrophy, which combined with enhanced pan-pTreg cells, can significantly contribute to age-related chronic inflammation in the elderly.

Protectin DX restores Treg/Th17 cell balance in rheumatoid arthritis by inhibiting NLRP3 inflammasome via miR-20a

Shengwei Jin et al.

Summary: Protectin DX (PDX) was found to effectively ameliorate the progression of collagen-induced arthritis by restoring Treg/T(h)17 cell balance, and this effect was mediated by inhibiting the NLRP3 inflammasome pathway via miR-20a.

IL-6 regulates the bone metabolism and inflammatory microenvironment in aging mice by inhibiting Setd7

Jiwei Wang et al.

Summary: The study reveals that IL-6 activates the TLR2, TLR4, and AKT pathways, inhibits the expression of beta-catenin and Setd7, and promotes proinflammatory gene expression in BMSCs. Setd7 expression in bone tissues of aged mice is suppressed, but it can promote BMSC osteogenic differentiation.

The elevated level of IL-1? in the bone marrow of aged mice leads to MSC senescence partly by down-regulating Bmi-1

Xueling Zheng et al.

Summary: Osteoporosis is more prevalent as individuals age due to bone marrow mesenchymal stem cells (MSCs) undergoing senescence, resulting in reduced bone formation and repair. Studies showed that MSCs from aged mice exhibited senescent phenotypes with decreased Bmi-1 expression and increased IL-1β levels in the bone marrow microenvironment, leading to MSC senescence.
