Motile cilia genetics and cell biology: big results from little mice

Lance Lee et al.

Summary: Over the past two decades, our understanding of motile cilia and their role in disease has greatly increased, with critical information and insight coming from the analysis of mouse models. Mouse models have made significant contributions to genetic and cell biological studies of motile cilia, and have been at the forefront of research on mammalian motile cilia and PCD pathogenesis.

Bi-allelic mutations in MCIDAS and CCNO cause human infertility associated with abnormal gamete transport

Cong Ma et al.

Summary: Mutations in MCIDAS and CCNO genes cause infertility by reducing the number of multiciliated cells in the efferent ducts/oviducts. In vitro fertilization/intracytoplasmic sperm injection interventions were successful in treating patients with these mutations, indicating a potential treatment option for infertility caused by reduced generation of multiple motile cilia.

Motile cilia and airway disease

Marie Legendre et al.

Summary: The article discusses the role of regulated airway epithelial development in the differentiation of motile ciliated cells in the human respiratory tract, as well as ciliary dysfunction caused by genetic mutations. Advances in understanding respiratory epithelial development and ciliary biology have the potential to improve disease diagnosis, management, and family counseling.

Motility of efferent duct cilia aids passage of sperm cells through the male reproductive system

Isabella Aprea et al.

Summary: Recent studies have shown that the motility of efferent duct cilia is crucial for male reproductive fitness in both mice and humans. Deficiency of the DNAH5 gene can lead to obstructive oligozoospermia, affecting male fertility but not sperm motility. Further research on environmental factors or genes causing efferent duct cilia dysmotility may provide insights into other causes of oligozoospermia.

Genetic Background and Clinicopathologic Features of Adult-onset Nephronophthisis

Takuya Fujimaru et al.

Summary: This study analyzed the genetic background and clinicopathologic features of adult NPH in 18 sporadic adult patients, finding that thick TBM duplication is a specific finding in adult NPH. Patients with pathogenic mutations were significantly younger, and older patients tended to have no pathogenic mutations even when suspected to have NPH by renal biopsy.

Expanding the genetic landscape of oral-facial-digital syndrome with two novel genes

Alanna Strong et al.

Summary: Oral-facial-digital syndromes (OFDS) are a rare group of Mendelian disorders characterized by developmental abnormalities of the oral cavity, face, and digits caused by dysfunction of the primary cilium. These syndromes can be inherited in X-linked dominant, X-linked recessive, and autosomal recessive manners. Despite many causal genes being identified, up to 40% of OFDS cases have unknown genetic basis.

Whole genome sequencing in the diagnosis of primary ciliary dyskinesia

Gabrielle Wheway et al.

Summary: The study revealed that genetic testing plays a crucial role in diagnosing primary ciliary dyskinesia (PCD), especially in cases with atypical disease history. Whole genome sequencing (WGS) is an effective approach that can detect variants missed by prior gene panel testing and is a powerful tool for discovering novel genes.

CEP164 is essential for efferent duct multiciliogenesis and male fertility

Mohammed Hoque et al.

Summary: The study concludes that male infertility in mice lacking CEP164 is caused by sperm agglutination and obstruction of efferent ducts due to loss of multicilia. This highlights the essential role of the distal appendage protein CEP164 in male fertility.

Progress in Diagnosing Primary Ciliary Dyskinesia: The North American Perspective

Michael Glenn O'Connor et al.

Summary: Primary Ciliary Dyskinesia (PCD) is a rare disease that affects respiratory ciliary function, with diagnosis requiring nasal nitric oxide (nNO) testing, genetic testing, or ciliary electron micrograph analysis. Guidelines recommend referral to a PCD Foundation accredited center for patients with compatible clinical phenotypes.
