Communication between cells: exosomes as a delivery system in prostate cancer

Pia Giovannelli et al.

Summary: Prostate cancer remains the second leading cause of cancer-related death in men despite advances in screening and diagnostic protocols. The mechanisms of disease progression are still under debate, with a focus on the role of communication between prostate tumor cells and the surrounding microenvironment. Exosomes released by prostate cancer cells play a critical role in intercellular communication and research into their role in malignancy and progression is ongoing.

The androgen receptor/filamin A complex as a target in prostate cancer microenvironment

Marzia Di Donato et al.

Summary: Prostate cancer-associated fibroblasts express a transcriptional-incompetent androgen receptor which, upon binding with filamin A, mediates migration and invasiveness. These fibroblasts induce an increase in prostate cancer organoid size by moving towards epithelial prostate cancer cells. Androgen enhances these effects through the assembly of an androgen receptor/filamin A complex in cancer-associated fibroblasts.

Therapeutic potential of TRPM8 antagonists in prostate cancer

Marzia Di Donato et al.

Summary: TRPM8 antagonists can inhibit androgen-dependent prostate cancer cell proliferation, migration, and invasiveness, and have a significant reverse effect on the spheroid size increase induced by androgens in prostate cancer cells. Selected antagonists interfere with non-genomic androgen action, disrupt the androgen-induced androgen receptor/TRPM8 complex assembly, and prevent the increase in intracellular calcium levels in prostate cancer cells.

ING Tumour Suppressors and ING Splice Variants as Coregulators of the Androgen Receptor Signalling in Prostate Cancer

Anna Melekhova et al.

Summary: ING proteins have been identified as potential drug targets in prostate cancer research, playing a role in inhibiting cell growth and tumor development. Their splice variants offer a novel direction for investigating the biology of ING factors in various malignancies.

Dissecting the Hormonal Signaling Landscape in Castration-Resistant Prostate Cancer

Fabrizio Fontana et al.

Summary: Understanding the molecular mechanisms of prostate cancer progression to the castration-resistant stage is crucial for improving therapeutic options. The activation of the androgen/androgen receptor axis and the GnRH/GnRH-R axis in CRPC cells play significant roles in antitumor activity, suggesting potential therapeutic implications.
