Levomepromazine and clozapine induce the main human cytochrome P450 drug metabolizing enzyme CYP3A4

Przemyslaw J. Danek et al.

Summary: The study found that levomepromazine and clozapine can induce the expression of CYP3A4 in human hepatocytes, but do not have a significant effect on the activity of other CYP enzymes. Further in vivo studies are needed to confirm the induction of CYP3A4 by these drugs and to assess their impact on drug metabolism.

Summary: Excess cholesterol accumulation has been shown to play a critical role in the pathogenesis of multiple diseases, including liver diseases, diabetes, and osteoporosis. The ubiquitous toxicity induced by excess cholesterol may potentially expand the therapeutic use of cholesterol-lowering medications.

Activation of pregnane X receptor induces atherogenic lipids and PCSK9 by a SREBP2-mediated mechanism

Mikko Karpale et al.

Summary: This study investigated the role of PXR receptor in regulating atherogenic lipid levels in humans and found that PXR activation induces cholesterol synthesis, leading to elevated LDL and total cholesterol. The PXR-SREBP2 pathway was identified as a novel regulator of cholesterol and PCSK9 synthesis, providing a molecular mechanism for drug- and chemical-induced hypercholesterolemia.

Effects of proprotein convertase subtilisin/kexin type 9 and nilotinib plasma concentrations on nilotinib-induced hypercholesterolaemia in patients with chronic myeloid leukaemia

Maiko Abumiya et al.

Summary: This study investigated the relationship among nilotinib plasma trough concentration, LDL cholesterol, and PCSK9 plasma concentration in patients with chronic myeloid leukaemia. Results showed that LDL cholesterol concentrations increased significantly after nilotinib treatment, with higher PCSK9 concentrations at 3 months post-treatment. Patients with a higher change rate in PCSK9 had a significantly higher change rate in LDL cholesterol, suggesting that nilotinib may lead to hypercholesterolaemia by increasing PCSK9 concentrations.
