Addition of immunotherapy to chemotherapy for metastatic triple-negative breast cancer: A systematic review and meta-analysis of randomized clinical trials

Xingfa Huo et al.

Summary: This study showed that adding PD1/PD-L1 blockade to chemotherapy improved progression-free survival in PD-L1 positive metastatic triple-negative breast cancer patients, as well as in the intention-to-treat population. However, no significant benefit in overall survival was observed in patients regardless of PD-L1 status or intention-to-treat population after adding PD1/PD-L1 blockade. The combination of immunotherapy and chemotherapy was associated with a higher rate of adverse events.

xCT-Driven Expression of GPX4 Determines Sensitivity of Breast Cancer Cells to Ferroptosis Inducers

Namgyu Lee et al.

Summary: The study revealed that breast cancer cells are more sensitive to inducers of ferroptosis such as Erastin and Rsl-3 due to their dependency on the xCT/GPX4 axis. This finding provides a strategic basis for targeting the anti-ferroptotic machinery of breast cancer cells based on their xCT status.

Elzbieta Cecerska-Heryc et al.

Summary: Reactive oxygen species (ROS) are significant in human cells, with antioxidant enzymes playing a crucial role in maintaining balance and diagnosing neoplastic diseases. Lowered activity of SOD, CAT, and GPx is characteristic of various cancers, making SOD, CAT, and XOR promising prognostic markers in lung, bladder, ovarian, and colon cancer.

A p97/Valosin-Containing Protein Inhibitor Drug CB-5083 Has a Potent but Reversible Off-Target Effect on Phosphodiesterase-6

Henri Leinonen et al.

Summary: CB-5083, an inhibitor of p97/valosin-containing protein (VCP), initially failed in cancer treatment due to adverse effects on vision, but shows potential for treating neuromuscular VCP disease. Research indicates that CB-5083 has a reversible inhibitory effect on retinal photoreceptor function, without long-term consequences on retinal function or structure.

p97/VCP is highly expressed in the stem-like cells of breast cancer and controls cancer stemness partly through the unfolded protein response

Chuang Li et al.

Summary: p97, an ATPase commonly overexpressed in many cancers, is positively correlated with histological grade, tumor size, and lymph node metastasis in breast cancers. High levels of p97 are found in stem-like cancer cells and cancer stem cell populations, with p97 expression positively correlating with SOX2. Depletion or inhibition of p97 leads to reduced cancer growth and CSC population, primarily affecting ALDH(+) CSCs and CSC-enriched mammospheres.
