Radiomic machine learning for predicting prognostic biomarkers and molecular subtypes of breast cancer using tumor heterogeneity and angiogenesis properties on MRI

Ji Young Lee et al.

Summary: This study investigated machine learning approaches for radiomics-based prediction of prognostic biomarkers and molecular subtypes of breast cancer using MRI, with texture and perfusion analyses. Texture and perfusion parameters were associated with hormone receptors, Ki67, grade, and molecular subtypes. The random forest model integrating texture and perfusion parameters showed the highest performance, with important parameters for prediction being texture irregularity (entropy) and relative extracellular extravascular space (V-e).

Dynamic contrast-enhanced breast MRI features correlate with invasive breast cancer angiogenesis

Jennifer Xiao et al.

Summary: The study evaluated the association of breast cancer features on dynamic contrast-enhanced MRI with MVD, a marker for angiogenesis. Lesions with high MVD levels showed higher peak SER and WF, and some radiomics texture features were promising in predicting increased MVD. DCE-MRI can non-invasively assess breast cancer angiogenesis, which could help optimize treatments based on tumor biology.

Summary: This study explored the use of nanoradiomics to differentiate tumors based on TAM burden, finding that conventional tumor metrics were ineffective in distinguishing tumors with varying TAM burden. However, machine learning-augmented nanoradiomic analysis revealed texture differences that enabled the differentiation of low and high TAM tumors.

Predicting local tumour progression after ablation for colorectal liver metastases: CT-based radiomics of the ablation zone

F. C. R. Staal et al.

Summary: The study aimed to investigate whether CT-based radiomics of the ablation zone (AZ) can predict local tumour progression (LTP) after thermal ablation for colorectal liver metastases (CRLM). Results showed that combining clinical features and radiomics features in a combined model yielded a better prediction of LTP compared to using radiomics alone. CT-based radiomics of the AZ and periablational rim (PAR) may have the potential to aid in predicting LTP during follow-up in patients with CRLM.

Non-invasive measurement of PD-L1 status and prediction of immunotherapy response using deep learning of PET/CT images

Wei Mu et al.

Summary: The study suggests that quantitative imaging can serve as an alternative to immunohistochemistry for predicting PD-L1 expression status in NSCLC patients treated with immune checkpoint inhibitors. The developed deeply learned score was able to accurately predict durable clinical benefit, progression-free survival, and overall survival in retrospective and prospective cohorts of patients. The findings support the use of this score as a surrogate for PD-L1 measurement in guiding individual treatment decisions.
