Angiopep-2-functionalized nanoparticles enhance transport of protein drugs across intestinal epithelia by self-regulation of targeted receptors

Summary: The study demonstrated that modifying nanoparticles with angiopep-2 (ANG) targeting LRP-1 on the intestine can enhance both apical endocytosis and basolateral transcytosis. The increased transcytosis efficiency of ANG NPs led to significant improvement in insulin absorption and blood glucose reduction in diabetic rats. This approach of self-regulating receptor distribution on polarized intestine cells shows promise for the design of oral delivery systems for protein/peptide drugs.
