Models of Idiosyncratic Drug-Induced Liver Injury

Tsuyoshi Yokoi et al.

Summary: Drug-induced liver injury (DILI) is a major concern during drug development and post-marketing, with both intrinsic and idiosyncratic classifications. Predicting idiosyncratic DILI, associated with severe liver damage, is challenging due to its low incidence and complex nature. Animal models have been used to study DILI mechanisms and predict potential risks.

An in vitro coculture system of human peripheral blood mononuclear cells with hepatocellular carcinoma-derived cells for predicting drug-induced liver injury

Shingo Oda et al.

Summary: This study established a coculture system using PBMCs and HepG2 cells and selected biomarkers that can predict DILI risk. A stepwise logistic regression model using the 12 identified genes showed the highest predictive performance with a high sensitivity/specificity.

Characterization of plasma protein binding in two mouse models of humanized liver, PXB mouse and humanized TK-NOG mouse

Maki Miyamoto et al.

Summary: The unbound fractions in plasma in two humanized liver mouse models, PXB and humanized TK-NOG mice, were compared with human values. PXB mice showed a good relationship with humans, while humanized TK-NOG mice showed a weak correlation. Different profiles of plasma protein binding were observed between the two mouse models, indicating the need for f(up) evaluation in each model for pharmacological studies.

Combining In Vivo Data with In Silico Predictions for Modeling Hepatic Steatosis by Using Stratified Bagging and Conformal Prediction

Summary: In this study, reliable in silico models were developed for predicting hepatic steatosis by combining various descriptor combinations, leading to improved model performance. Both meta-classifiers and the conformal prediction framework were found to be useful for handling imbalanced toxicity data sets.

Characterizing the reproducibility in using a liver microphysiological system for assaying drug toxicity, metabolism, and accumulation

Andres Rubiano et al.

Summary: The study characterized the reproducibility of liver microphysiological systems (MPSs) in predicting hepatic drug effects through experiments on drug toxicity, metabolism, and intracellular accumulation. The results demonstrated that the liver MPS can reliably produce consistent experimental outcomes for drug evaluation applications, showing better stability and functionality compared to other culture platforms. The study also provided general considerations and recommendations for using liver MPSs in drug development.

Advancements in stem cell-derived hepatocyte-like cell models for hepatotoxicity testing

Meixian Jin et al.

Summary: Drug-induced liver injury is a major cause of clinical trial failures and high rates of drug attrition. Stem cells are considered stable sources for drug hepatotoxicity screening models due to their self-renewable ability and multipotent competence. 3D culture, which can better mimic the in vivo microenvironment than 2D culture, is commonly used for hepatocyte culture and drug screening.

High-Content Screening for the Detection of Drug-Induced Oxidative Stress in Liver Cells

Maria Teresa Donato et al.

Summary: Drug-induced liver injury (DILI) is a major obstacle in drug development, and understanding the mechanisms behind it is crucial for better drug design. Oxidative stress leading to oxidative damage is one of the main mechanisms involved in DILI. High-content screening technology is used to identify safety risks and is an effective tool for pre-lethal mechanism detection.

Drug properties and host factors contribute to biochemical presentation of drug-induced liver injury: a prediction model from a machine learning approach

Andres Gonzalez-Jimenez et al.

Summary: The presentation of drug-induced liver injury (DILI) varies, with some drugs showing consistent injury patterns while others result in broader types of liver damage. Factors including drug properties and host factors play a role in determining the type of DILI. By considering both drug and host factors, it is possible to differentiate hepatocellular from cholestatic damage with reasonable accuracy, aiding in future DILI research and causality assessment.

Summary: Idiosyncratic drug reactions are unpredictable adverse reactions, with idiosyncratic drug-induced liver injury being the most serious consequence. Research on mouse models suggests that immune checkpoint inhibitors may increase the risk of drug-induced liver injury with specific medications, but can also reduce liver injury in certain cases.

Summary: This study identified factors such as drug metabolism, serum bilirubin, and ALP levels at DILI onset that influence the recovery time for DILI patients. Delayed recovery predictions may warrant additional investigations and longer follow-ups for patients.

Summary: This study confirmed the role of adaptive immune response in drug-induced liver injury (DILI) by analyzing leukocyte populations and immune checkpoint expressions. Results showed that DILI patients had higher levels of activated T cells and intracellular CTLA-4 at the initial phase, approaching those of healthy liver controls over time. Increased expression of negative immune checkpoints in DILI patients may suggest peripheral tolerance regulation.
