Thioether-Based Polymeric Micelles with Fine-Tuned Oxidation Sensitivities for Chemotherapeutic Drug Delivery

Summary: Oxidation-sensitive drug delivery systems have the potential to improve the efficacy and safety of chemotherapy, but the selectivity between cancer cells and healthy cells remains a challenge. This study reports the design and characterization of polymeric micelles containing different oxidation-sensitive thioether groups. The micellar structure becomes hydrophilic upon thioether oxidation, leading to destabilization. The results show that the oxidation reaction can be modulated by changing the substituent groups on the sulfur atom. These micelles have the potential to improve therapeutic efficacy and safety in cancer treatment.

Phosphorylcholine-Grafted Molecular Bottlebrush-Doxorubicin Conjugates: High Structural Stability, Long Circulation in Blood, and Efficient Anticancer Activity

Summary: In this study, a poly(methacryloyloxyethyl phosphorylcholine) (pMPC)-grafted molecular bottlebrush (PCMB) conjugated with a cancer drug, doxorubicin (DOX), showed better antitumor efficacy compared to a pMPC-based linear polymer (LP) conjugated with DOX. The improved efficacy of PCMB-DOX was attributed to the lack of agglomeration and limited release of DOX at the tumor site, unlike LP-DOX which formed agglomerates and compromised DOX release. These findings highlight the potential of molecular bottlebrushes as building blocks for drug carriers in the design of polymer-based nanomedicines.

Cascade Reactions by Nitric Oxide and Hydrogen Radical for Anti-Hypoxia Photodynamic Therapy Using an Activatable Photosensitizer

Summary: In this study, a unimolecular glutathione and light coactivatable photosensitizer targeting mitochondria was developed, demonstrating high performance PDT effect through type I, type II reactions, and secondary radicals-participating reactions. With a relatively large two-photon absorption cross section for photoexcitation, the new photosensitizing system shows promising applications in both normoxic and hypoxic environments.
