FDAApproval Summary: Olaparib Monotherapy or in Combination with Bevacizumab for the Maintenance Treatment of Patients with Advanced Ovarian Cancer

Shaily Arora et al.

Summary: FDA approved olaparib monotherapy and combination therapy with bevacizumab for the first-line treatment of advanced ovarian cancer with BRCA mutations and homologous recombination deficient-positive status. Both treatments demonstrated clinically meaningful improvements in progression-free survival and favorable benefit-risk profiles in clinical trials.

Hypoxia and the phenomenon of immune exclusion

Violena Pietrobon et al.

Summary: Cancer immunotherapy is considered a promising strategy against many types of cancer, but the exclusion of lymphocytes from the tumor nest limits its efficiency in solid tumors. Hypoxia, a hallmark of most solid tumors, may play an upstream role as a common biological determinant of immune exclusion in solid tumors.

Sevoflurane and Desflurane Exposure Enhanced Cell Proliferation and Migration in Ovarian Cancer Cells via miR-210 and miR-138 Downregulation

Summary: The study showed that sevoflurane and desflurane enhanced ovarian cancer cell malignancy by deactivating miRNAs and increasing HIF-1 alpha expression. Further research is needed to determine the translational value of these findings.

The ER stress response mediator ERO1 triggers cancer metastasis by favoring the angiogenic switch in hypoxic conditions

Ersilia Varone et al.

Summary: Deficiency of ERO1 inhibits tumor cell migration and lung metastases by negatively regulating tumor angiogenesis and impeding oxidative folding of angiogenic factors; ERO1 levels are higher in aggressive basal breast tumors and correlate inversely with disease- and metastasis-free interval, suggesting its potential as a marker for tumor aggressiveness.

Stanniocalcin 1 is a phagocytosis checkpoint driving tumor immune resistance

Heng Lin et al.

Summary: Immunotherapy can induce durable clinical responses in some cancer patients, but resistance poses a major challenge. The expression of tumor STC1 is correlated with immunotherapy efficacy and negatively associated with patient survival across different cancer types. This study suggests that tumor STC1 may inhibit APC phagocytosis and contribute to tumor immune evasion and immunotherapy resistance.

Targeting stanniocalcin-1-expressing tumor cells elicits efficient antitumor effects in a mouse model of human lung cancer

Kotaro Abe et al.

Summary: The study investigated the potential of targeting STC-1 in tumor cells for gene therapy in lung cancer, confirming the important role of STC-1 in lung cancer cells. The findings suggest that conferring cytotoxicity in STC-1-positive lung cancer cells using a suicide gene may be a promising therapeutic strategy for lung cancer.

Gold Nanoparticles Affect Pericyte Biology and Capillary Tube Formation

Sasikarn Looprasertkul et al.

Summary: The study revealed the effects of AuNPs on pericytes, showing inhibition of proliferation, decreased migration, and reduced PDGFR-β mRNA expression. Ultrastructural analysis showed the presence of AuNPs in late endosomes, autolysosomes, and damaged mitochondria in pericytes. In 3D capillary tube formation, AuNPs led to abnormal positioning of pericytes, resulting in incomplete tube formation.

Estrogen/ER in anti-tumor immunity regulation to tumor cell and tumor microenvironment

Tiecheng Wang et al.

Summary: Estrogen and its receptor play crucial roles in regulating autoimmune diseases and anti-tumor immunity. The mechanisms of estrogen-induced immunity, including canonical and non-canonical signal pathways, are important for understanding tumor immunity and providing potential targets for cancer immunotherapy.

Extracellular Vesicle Transmission of Chemoresistance to Ovarian Cancer Cells Is Associated with Hypoxia-Induced Expression of Glycolytic Pathway Proteins, and Prediction of Epithelial Ovarian Cancer Disease Recurrence

Mona Alharbi et al.

Summary: Ovarian cancer is one of the most lethal cancers worldwide affecting women, with non-specific early symptoms. sEVs produced under low oxygen tension can induce a metabolic switch in ovarian cancer cells, promoting resistance to carboplatin. The presence of glycolysis pathway proteins in sEVs is closely associated with ovarian cancer recurrence.

Normalizing Tumor Vasculature to Reduce Hypoxia, Enhance Perfusion, and Optimize Therapy Uptake

Kathy Matuszewska et al.

Summary: Solid tumors require rapid development of new blood vessels to support their metabolic needs, which can lead to tumor hypoxia and a pro-tumorigenic environment. Targeting tumor vasculature normalization can reduce hypoxia and enhance therapy efficacy. Understanding tumor angiogenesis biology and exploring new clinical approaches, including combination therapies with immunotherapy, are crucial to advancing vascular targeting as a therapeutic strategy.
